Objective: Measure the PUFAs content in Oviductus Ranae in different periods and production areas in Jilin Province,and perform cluster analysis to determine the best harvesting period and production area of Oviductus Ranae;explore the correlation between gene expression and PUFAs content,and cultivate it And harvest to provide a basis.Methods: Establish an in vitro analytical method for the determination of polyunsaturated fatty acids(PUFAs)in Oviductus Ranae;Gas chromatography was used to analyze the six annual growth periods(sting stage,spawning period,predation period,stinging period,scattered hibernation period and social hibernation period)in Jingyu County,Jilin Province,and six Rana chensinensis habitats in Jilin Province(Total PUFAs,palmitic acid(C16:0),stearic acid(C18:0),C18:ln9,C18 in the fallopian tube of Rana chensinensis collected from Antu,Wang Qing,Huadian,Jingyu,Linjiang and Jiaohe): Determination of the content of 3n3,AA,EPA and DHA,using real-time fluorescent quantitative PCR(RT-PCR)to synthesize PUFAs related to fatty acid dehydrogenase ?6 fatty acid dehydrogenase(?6-FAD),?5 fatty acid dehydrogenation The gene expression levels of enzymes(?5-FAD),?9 fatty acid dehydrogenase(?9-FAD),and n-3 fatty acid dehydrogenase are determined,and the fatty acid content and related key enzyme genes obtained in different periods and production areas are determined The expression quantity is analyzed separately for regularity and clustering,and then SPSS software and R language software are used to combine the two for correlation analysis.Results: In different periods,the content of PUFAs in the fallopian tubes of Rana chensinensis was the highest during the scattered hibernation period,followed by the stinging period and the gregarious hibernation period,and the lowest during the stinging period,the oviposition period and the predation period;among different habitats,Jingyu China Forest The comprehensive content of PUFAs in the fallopian tubes of frogs is higher,followed by the producing areas Wangqing,Huadian and Linjiang,and the lowest producing areas Antu and Jiaohe.C18:1n9 and EPA have important reference value in the identification of Oviductus Ranae collection period;C18:3n3 and DHA have important reference value in the identification of Oviductus Ranae production area;Expression of ?6-FAD and ?5-FAD genes It has an important reference value in the identification of the harvesting period of Oviductus Ranae;the expression of ?6-FAD and ?9-FAD genes has an important reference value in the identification of Oviductus Ranae from different producing areas.Conclusion: There are significant differences in the content of PUFAs in Hama oil from different regions and different harvest periods;the expression of key enzyme genes for PUFAs synthesis is strongly correlated with the content of PUFAs,and the quality of Hama oil can be further analyzed and evaluated through research.
