ObjectiveTo compare the expression of Interleukin-14 alpha(IL-14 α)in tumor tissue and normal tissue of B cell non-Hodgkin’s lymphoma(B-NHLs)to understand the relationship between the expression of Il-14 α and the curative effect and prognosis.Method1.Analysis of the difference between human B cell lymphoma and normal tissue using GEPIA database;2.40 cases of primary Diffuse large B-cell lymphoma(DLBCL)from August 2016 to December 2019 were collected from the North Jiangsu People’s Hospital,the expression of IL-14α in frozen tissues of lymphoma patients was detected by q RT-PCR,the expression of IL-14α in serum of 16 patients before and after treatment was detected by Western Blot,and the expression of IL-14α was analyzed by Graph Pad 8.0;3.Patients were followed up to analyze the relationship between IL-14α expression level and efficacy.Kaplan-Meier method was used to analyze the survival curve of the patients.Log-rank univariate test was performed to analyze the relationship between IL-14α expression and the survival prognosis of the patients.Receiver operating characteristic curve(ROC curve)was used to analyze the relationship between IL-14α and recurrence risk.Result1.Based on the analysis of GEPIA database,we found that the expression level of IL-14α in DLBCL tumor tissues(n=47)was significantly higher than that in normal tissues(n=337),and the difference between the two was statistically significant(P < 0.05).Subsequently,we further analyzed the relationship between the expression level of IL-14α and the overall survival rate of DLBCL patients,and the results showed that the prognosis of patients with high IL-14α expression level was worse than that of patients with low IL-14α expression level,although there was no significant statistical significance between the two,there was a certain trend of difference.2.In this study,a total of 40 DLBCL tumor tissue samples were collected,and the expression of IL-14α in the tissue samples was detected by q RT-PCR.According to the results of the recent efficacy evaluation,the patients were divided into CR group(n=24)and PR/SD/PD group(n=16).The expression level of IL-14α in the two groups was compared,and the results showed that the initial expression level of IL-14α in the CR group was significantly lower than that in the PR/PD/SD group(P <0.05).3.In 40 DLBCL patients,the PFS of low IL-14α expression was 83% vs 57%(P = 0.19),and the OS of 5 years was 92% and 64%(P=0.06),respectively,but there is a more obvious trend of difference.4.The ROC curve was drawn and the area under curve(AUC)was calculated according to the recurrence of 40 DLBCL patients after standard treatment.The AUC value of IL-14α was 0.742,P=0.01,95%CI(0.591,0.894).When the optimal critical value was 2.4,the Youden index was the highest,and the sensitivity and specificity were 72.2% and 68.2%,respectively.5.Serum levels of IL-14 α were measured by Western-blot in 16 patients with B-NHLs before and after first-line therapy,including 3 patients with DLBCL(18.8%)and 13 patients with other b-cell lymphoma(81.3%),the results showed that the level of serum IL-14 α in B-NHLs after treatment was significantly lower than that before treatment(p = 0.04).The serum levels of IL-14α in 16 patients after treatment were down-regulated in 8 patients(75%),up-regulated in 8 patients(75%)and 25%(25%).The PFS of IL-14α down-regulated group and up-IL-14α down-regulated group were 75% vs 37.5%,respectively,the median follow-up period was 32 months,the difference was statistically significant(p = 0.02).Conclusion1.IL-14α was highly expressed in DLBCL lymphoma tissues compared with normal tissues.2.The expression level of IL-14α and its changes after treatment may be related to the prognosis,efficacy and recurrence risk of patients,which is worthy of further study.
