This thesis is consisted of three chapters.Firstly,the chemical constituents of Pholidota articulata were studied.Then the content of three phenanthrenes and two bibenzyls from chloroform fractions of this plant were determinated by using HPLC.Finally,the content of 16 mineral elements in the roots,stems,leaves,flowers and fruits of this plant were determinated by inductively coupled plasma optical emission spectrometry(ICP-OES).Objective:This study systematically investigated the chemical constituents of P.articulata Lindl.and mainly monitored the level of heavy metals in roots,stems,leaves,flowers and fruits of this plant to consider its health risk.Methods:(1)Silica gel,sephadex LH-20 gel column chromatographic coupled with recrystallization techniques were used to separate and purify the chemical constituents.Their structures were elucidated based on standard,NMR spectroscopic and reported data.Among these monomer compounds,five isolated compounds were used as standard for HPLC analysis.(2)Mineral elements were determined by ICP-OES after microwave digestion with HNO3-H2O2(5:3,v/v)as the digestion solvent,and the statistical techniques were performed to investigate relationships in data.Furthermore,the estimated daily intake(EDI),target hazard quotients(THQ)and hazard index(HI)were calculated to evaluate the health risk assessment.Results:(1)A total of 23 compounds were yielded from ethyl acetate fractions of P.articulata Lindl.and identified as flavidin(1),flaccidin(2),imbricatin(3),2,6-dimethoxy-9,10-dihydro-5H-phenanthro[4,5-bcd]pyran(4),coelonin(5),lusianthridin(6),hircinol(7),gigantol(8),batatasin III(9),cirrhopetalidin(10),cis-3,3′-dihydroxy-5-methoxystibene(11),β-sitosterol(12),stigmasterol(13),glut-5-en-3-ol(14),scopoletin(15),scoparone(16),7,8-dimethoxy coumarin(17),trans-cinnamic acid(18),4,4′-dihydroxy-diphenylmethane(19),laurostearic acid(20),hydroxyphenyl propionate(21),3-methoxybenzaldehyde(22).(2)The analysis method of five target compounds(1,5,6,9,10)was validated and demonstrated that all calibration curves were presented excellent linear regressions with keeping 0.9999of R2.Their structures were very stable for 48 h.The resolutions were more than 1.50by comparing the adjacent peaks.Symmetry factors were between 0.95 and 1.05.The average recoveries lay between 91.5%and 102.9%.(3)The content of 16 mineral elements in the roots,stems,leaves,flowers and fruits of P.articulata Lindl.were different.After statistical analysis,a significant or an extremely significant correlation was yielded from the original data,and five organs were clustered reasonably into three groups and could include ten characteristic heavy metals.And it was important to emphasize that the EDI for any of the heavy metals(except Al)was far lower than the oral reference dose(RFD),THQ were less than 1,the HI of five organs were followed by root(7.24)>flower(3.63)>leaf(1.90)>stem(0.71)>fruit(0.36).Conclusion:(1)The systematical investigation on the chemical constituents of P.articulata Lindl.have obtained seven phenenthrenes,three bibenzyls,three coumarins and nine other compounds.Moreover,compounds 3,4,7,10,11,13~17,19~22were first reported from the whole of this plant,whereas six compounds(10,14~17and 22)were first recorded in the genus Pholidota.(2)The developed HPLC method can simultaneously separate and determine of five compounds from chloroform fractions in different plants.On the other hand,this is the first report on the validation of a method for the quantitative analysis of flavidin(1)and cirrhopetalidin(10).(3)The ICP-OES method can simultaneously determine the contents of 16 mineral elements in different organs of P.articulata Lindl.The statistical analysis could be effectively applied in data analysis of mineral elements.Given the toxic health risk of the heavy metals,we found the stem and fruit of this plant can be edible for a long time.This work may provide a scientific reference for evaluating the possibility of potential nutritional and pharmaceutical value.
