| ObjectiveThe model of heart failure induced by Transverse aortic constriction in mice was established.The success of the model and the characteristic changes of cardiac structure and function were evaluated by high frequency echocardiography and other laboratory methods.MethodsThirty-six C57 BL / 6J male mice were randomly divided into three groups: control group(SHAM group,n = 12)1 day before operation,group 1 week after operation(TAC1 group,n = 12),group 6 weeks after operation(TAC2 group,n = 12).TAC1 and TAC2 groups were treated with Transverse aortic constriction to establish pressure overload heart failure model.Left ventricular posterior wall thickness(LVPW T),left ventricular diameter,left ventricular volume,left ventricular systolic and diastolic function were measured by echocardiogr aphy at 1 day before operation and 1,6 weeks after operation.At 6 weeks after operation,weigh each heart and body weight in mice,calculate the heart weight ratio;Masson staining and hematoxylin eosin staining were used to observe myocardial histopathological changes;Detect the protein expression of ANP which is a sign of myocardial hypertrophy by Western blot.ResultsEchocardiographic data showed that at 1 week after operation,the left ventricular wall thickness in TAC1 group increased compensatory,left ventricular systolic function was normal and showed compensatory increase,left ventricular diastolic function decreased;the left ventricular diameter and volume in TAC2 group were increased compared with those in SHAM group and TAC1 group at 6 weeks after operation(P < 0.05),the indexes reflecting systolic function were significantly lower than those in the other two groups(P < 0.01),and the indexes of diastolic function in TAC1 group and TAC2 group were lower than those in SHAM group,especially in TAC2 group.At 6 weeks after operation,the heart weight(HW)and heart body ratio(HW / BW)of TAC2 group were higher than those of SHAM group(P < 0.05).HE and Masson staining showed TAC2 group myocyte hypertrophy,collagen fiber content increase.Western blot shows TAC2 ANP protein expression were higher than that of control group.ConclusionsHigh frequency echocardiography could show that mice heart failure in the process,characteristic changes of cardiac structure and function,for the transverse aortic constriction heart failure model,and the application of translational medicine in basic medicine,provide theoretical basis. |
