Salidroside Modulates PI3K/Akt/GSK-3β Signaling Pathway To Inhibit The Inflammatory Response Of Hyperglycemic Retinal Müller Cells In Rats

ObjectiveTo investigate the effect of salidroside(Sal)on the activation of Retinal Muller cells(r MC-1)cultured in high glucose(HG)in vitro,And the mechanisms involved in the PI3K/Akt/GSK-3β signaling pathwayMethodsr MC-1 was cultured in vitro,and the control group,HG group(glucose 50mmol/L),SAL group,HG+SAL(100,200,500 μM)group and HG+SAL+LY294002 group were set,PI3 K inhibitor.After 48 h of culture,the protein expression levels of IL-1β,TNF-α,IL-6 and the expression of NF-B p65 in the nucleus of each group were detected by Western blot and immunofluorescence.The m RNA levels of inflammatory cytokines(IL-1β,IL-6,IL-10)in r MC-1 were detected by real-time fluorescence quantitative PCRResultsWestern blot results showed that compared with the control group,the protein expression levels of p-Akt and p-GSK-3β in HG group were significantly decreased(P<0.05),and the protein expression levels of IL-1β,TNF-α and IL-6 in HG group were increased(P<0.05).Compared with HG group,the protein expressions of p-Akt and p-GSK-3β in HG+SAL group were significantly increased(P<0.05),and the protein expressions of IL-1β,TNF-α and IL-6 in HG+ SAL group were significantly decreased(P<0.05).Compared with HG+SAL group,the protein expression levels of p-Akt and p-GSK-3β in HG+Sal+LY294002 group were decreased(P<0.05),and the protein expression levels of IL-1β,TNF-α and IL-6 in HG+SAL+LY294002 group increased(P<0.05).The nucleoplasmic expression of NF-B p65 in HG group was detected by immunofluorescence.Compared with the control group,the expression of NF-B p65 in HG group was increased in the nucleus(P<0.05).Compared with the HG group(P<0.05),the expression of NF-B p65 in HG+SAL group tended to be expressed in cytoplasm.The expression of NF-B p65 in the nucleus of HG+Sal+LY294002 group was enhanced(P<0.05).Compared with the control group,the m RNA expression levels of IL-6 and IL-1β in HG group were significantly increased(P<0.05),and the m RNA expression of IL-10 in HG group was decreased(P<0.05).The expression level of anti-inflammatory cytokine IL-10 m RNA in HG+ SAL group(IL-6,IL-1β)decreased(P<0.05),and the expression level of anti-inflammatory cytokine IL-10 m RNA increased(P<0.05).The expression level of IL-1β m RNA was significantly increased(P<0.05),the expression level of IL-10 m RNA was decreased(P<0.05).ConclusionsSAL can reduce the inflammatory response in rMC-1 induced by HG,and one of the mechanisms may be the anti-inflammatory effect of SAL through PI3K/Akt/GSK-3β signaling pathway.
