Objective: To investigate the clinical efficacy and safety of neoadjuvant therapy of Anlotinib combined with chemotherapy in patients with limb refractory soft tissue sarcoma(STS).Methods: the data of patients with limb refractory STS who received preoperative neoadjuvant therapy with Anlotinib or chemotherapy alone from October2015 to January 2020 were retrospectively analyzed.The curative effect was evaluated according to RECIST 1.1 standard and the safety was evaluated according to CTCAE 4.0standard.The main evaluation criteria were objective response rate(ORR),disease progression free rate(PFR),limb salvage rate and safety at 3,6 and 12 months after operation.Results: a total of 30 patients were collected,including 10 patients in the combined chemotherapy group and 20 patients in the chemotherapy group.According to RECIST 1.1 criteria,partial response(PR,20% vs0),objective response rate(ORR,20%vs0),disease stability(SD,70% vs 60%),disease control rate(DCR,90% vs 60%);disease progression(PD,10% vs 40%);tumor change(-15.76±21.95%vs14.73±21.57%,P<0.05).PFR at 3,6 and 12 months postoperatively(100% vs 95%,88.89% vs 75% and77.78% vs 55%).Hematological and non-hematological toxicity were controllable.The most common grade 3 and 4 adverse events were leucopenia(40%vs40%,P>0.05)and neutropenic fever There were 30%vs30%,P>0.05),thrombocytopenia(30%vs25%,P>0.05),poor healing(30% vs5%,P>0.05),oral mucositis(10%vs0%,P>0.05),fatigue(30%vs35%,P>0.05)and hypertension(10%vs20%,P>0.05).Conclusion: according to our single center data,Anlotinib combined with doxorubicin and Ifosfamide has good efficacy and tolerability in the treatment of refractory STS patients.
