| ObjectiveTo explore the effect of fibroblast growth factor receptor inhibitor BGJ398 and in combination with rapamycin on the proliferation and apoptosis of ovarian cancer SKOV3 cells.MethodsThe ovarian cancer cell SKOV3 cells were cultured in vitio.The experimental group was treated with serial concentrations of BGJ398,rapamycin,or both inhibitors at a ratio of 1:1.The does range was 0.125-8×IC50of each compound.Cy Quant Cell Proliferation Assay Kit was used in the evaluation of cell growth.The apoptosis and cell cycle were evaluated using flow cytometry.ResultsBGJ398 alone or in combination with rapamycin could significantly inhibit the proliferation of the ovarian cancer SKOV3 cells in does-dependent manners,and joint efect of the two drugs had a higher inhibition rate(P<0.05),showing their synergistic effects.Flow cytometry revelated that BGJ398 alone or in combination with rapamycin could cause G0/G1 phase arrest,decreased cell number at S and G2 phase of ovarian cancer SKOV3 cells and induce apoptosis at the same time(P<0.05),showing their synergistic effects.Conclusions1.SKOV3 cells could be susceptible to co-inhibition of BGJ398 and rapamycin.2.BGJ398 and rapamycin can significantly inhibit the proliferation of the ovarian cancer SKOV3 cells,arrest cell cycle and induce cell apoptosis,and the two drugs administered in combination show synergistic effects. |
