Objective:To screen the differentially expressed proteins in the whole blood samples of Ewing sarcoma and to explore the molecular biological basis of the disease,so as to provide scientific basis for the research on the pathogenesis and development of Ewing sarcoma.Methods:i TRAQ was used to detect protein expression,and GO,KEGG and Western blotting were used to analyze differential proteins.Results:For i TRAQ,compared with healthy subjects,91 proteins were up-regulated and 36 were down-regulated in the prechemotherapy group.Compared with the group after chemotherapy,101 proteins were up-regulated and 35 were down-regulated in Ewing sarcoma group.In Ewing sarcoma,the expression was up-regulated compared with that of healthy people before chemotherapy,and the 8 proteins with the greatest difference were down-regulated after chemotherapy compared with those before chemotherapy:Myosin light chain2Alpha-actinin-2dunhao、Collagen alpha-1(I)chain、Superoxide dismutase 1、Annexin A6、Transgelin、Alpha-1-acid glycoprotein 1、Collagen alpha-2(VI)chain.Western blotting analysis was consistent with i TRAQ identification for the expression of SOD1.Conclusion:Superoxide dismutase 1 and other 7 proteins are associated with the occurrence and development of Ewing sarcoma.
