Objective:To study whether the proportion of regulatory T cell(Treg)in peripheral blood of beagle dogs increased before and after Aspirin-induced,and whether Treg promoted the migration ability and fibroblast differentiation of canine periodontal ligament stem cells(PDLSCs)after induction.Methods:Cultivate Beagle PDLSCs and identify their stem cell properties.Beagle dogs aged 8-10 months were selected,and Aspirin(75mg/day)was taken orally after 12 hours of fasting.Flow cytometry was used to detect the changes in the proportion of Treg cells in the peripheral blood of Beagle dogs before and after administration,and the Treg cells obtained before and after Aspirin induction Co-cultivation with Beagle PDLSCs,The migration ability of PDLSCs was detected by scratch experiment.Real-time quantitative polymerase chain reaction(RT-PCR)and enzyme-linked immunosorbent assay(ELISA)were used to detect Scleraxis(SCX)and type I collagen(COL)from m RNA and protein levels.-1)and basic fibroblast growth factor(b FGF)fibroblast factor index.Results:(1)Flow cytometry detected the increase in the ratio of Treg cells in the peripheral blood of Beagle dogs 3 hours after taking Aspirin.(2)Scratch experiment shows that Treg cells can promote the migration ability of PDLSCs after induction.(3)The results of RT-PCR and ELISA showed that the expressions of SCX,COL-1 and b FGF in the PDLSCs/Treg~+co-culture group were significantly higher than those in the PDLSCs group and were statistically significant(P<0.05).Conclusion:(1)Treg can promote the migration ability of PDLSCs after Aspirin induction(2)Treg can promote the expression of m RNA and protein of fibroblast-related genes COL-1,SCX and b FGF factor after Aspirin induction,and promote the differentiation of PDLSCs into the direction of fibrogenesis.
