Objective:This study aimed to explore the effect of COX-2 selective inhibitor(celecoxib)on adenomyosis and its relationship with cyclooxygenase-2(COX-2),vascular endothelial growth factor(VEGF),nerve growth factor(NGF),corticotropin-releasing hormone(CRH).Methods:By establishing a mouse model of adenomyosis and using celecoxib to treat adenomyosis,newly born female mice were randomly divided into control group,intervention group,and experimental group,with 6 mice in each group.After the model was established,they were reared for 3 months and collected the uterine tissues.hematoxylin-eosin(H&E)staining was used to observe the depth of endometrial infiltration of mouse adenomyosis.RT-PCR(reverse transcription PCR)and Western blot analysis were used to detect the expression of Cyclooxygenase-2(COX-2),Vascular endothelial growth factor(VEGF),Nerve growth factor(NGF),Corticotropin-releasing hormone(CRH)mRNA and protein in mice before and after celecoxib treatment.Results:(1)the uterus weight of mice aged 12 weeks in control group,intervention group and experimental group gradually increased(P=0.009,P=0.000,P=0.001),and the difference was statistically significant(P<0.05).(2)The endometrial infiltration depth of control group,intervention group and experimental group was compared in pairs(P=0.072,P=0.000,P=0.002),and the difference was statistically significant(P<0.05).The mRNA and protein expressions of COX-2,VEGF,NGF and CRH in intervention group and experimental group were higher than those in control group(P < 0.001),while the expressions of COX-2 and VEGF in intervention group were significantly lower than those in experimental group(P< 0.001),but there was no significant difference in NGF and CRH(P> 0.05).Conclusion:(1)A selective COX-2 inhibitor,celecoxib can reduce the degree of adenomyosis in mice.(2)COX-2,VEGF,NGF and CRH may be involved in the occurrence and development of adenomyosis.(3)Celecoxib is expected to be a potentially effective drug for the treatment of adenomyosis.
