| ObjectiveObjective to investigate the concentration of sST2 before and after the occurrence of aortic dissection in C57 BL / 6J mice.By studying the changes of sST2 level during the occurrence of aortic dissection,we can understand the relationship between sST2 and the occurrence of aortic dissection,in order to find new serum markers with high sensitivity and specificity,or establish a diagnostic model for clinical diagnosis It provides new objective evidence for the rapid diagnosis of aortic dissection.MethodThirty C57 BL / 6J male 3-week-old mice were selected and their body weights were measured before the test,and three groups,i.e.,control,BAPN and BAPN + Ang II groups,10 each,were randomly assigned in order of their body weights.Mice in the control group were given normal chow water,BAPN 0.5g/(kg.d)was added to the water of mice in the BAPN group,and BAPN was added to the water of mice in the BAPN +Ang II group 0.5 g /(kg.d),and were embedded subcutaneously at the dorsum of the neck before feeding in osmotic minipumps,which were filled with Ang Ⅱ,slowly injected at 1000 ng /(kg.min)speed,three groups of mice were fed for a total of 4 weeks,room temperature 21-23 ° C,relative humidity 50%-60%,and light exposure for 12 h per day.As mice died during feeding,the cause of death was analyzed immediately by dissection,whether it was lethal due to formation of an aortic dissection.All mice were sacrificed after 4 weeks,the aortas were removed at autopsy,the aortic diameters were measured,he staining and light microscopy were performed,the damage to the aortic wall was observed and photographed,and the incidence and mortality of aortic dissection in mice were calculated.Mouse serum was taken before feeding and after the mice died,and all blood samples were collected into blood collection tubes and centrifuged at 3000 rpm for 10 min at the clinical laboratory.The upper serum was quickly removed and stored at-80 ℃.Serum sST2 levels were detected with an ST2 assay kit(purchased from R & D systems,USA).Statistical analysis was performed using SPSS version 21.0software,comparisons between multiple groups were performed using one-way analysis of variance,receiver operating characteristic curve(ROC)was done,and the area under the curve(AUC)was calculated to analyze the correlation between sST2 and the occurrence of aortic dissection.Results1.Comparing the mortality of three groups,in the control group,the 28 day mortality was 0%;in BAPN group,the 28 day mortality was 10%;and in BAPN + AngⅡ group,the 28 day mortality was 100%.Compared with the control group and BAPN group,the incidence and mortality of aortic dissection rupture in BAPN + Ang Ⅱ group were significantly increased(P < 0.05).2.When comparing the incidence of aortic dissection between the three groups,no mice in the control group exhibited aortic dissection and the incidence of dissection was0%;8 mice in the BAPN group had their aortas dissected,and the incidence of dissection was 80%;all mice in the BAPN + Ang Ⅱ group were found to have aortic dissection and rupture after death,and the incidence of dissection was 100%,but there was no significant difference in the incidence of aortic dissection between BAPN group and BAPN + Ang Ⅱ group(80% vs.100%).3.Comparing the aortic wall thickness of three groups,BAPN + Ang Ⅱ group was significantly thicker than BAPN group(108.04 ± 29.53 μ m vs.71.17 ± 18.97 μ m,P <0.05)and control group(108.04 ± 29.53 μ m vs.47.86 ± 12.17 μ m,P < 0.05).4.There was no significant difference in the concentration of sST2 among the three groups before the experiment(P > 0.05).When sST2 concentrations in the plasma of mice from the three groups after death were compared,1.023 ng / ml in the control group,2.336 ng / ml in the BAPN group and 2.521 ng / ml in the BAPN + Ang II group.The concentrations of sST2 in the BAPN group and BAPN + Ang Ⅱ group were significantly higher than those in the control group(P < 0.05),but there was no significant difference between the BAPN group and BAPN + Ang Ⅱ group(P = 0.083).5.In ROC analysis,the area under the curve of sST2 for the diagnosis of aortic dissection was 0.889(95% CI: 0.742-1.000).ConclusionsThe results of the data from this experimental study showed that the concentrations of sST2 protein in plasma were significantly increased when aortic dissection or aortic wall injury occurred,and that the diagnostic value of sST2 protein for aortic dissection was high,and sST2 protein may be used as a novel biological marker for the diagnosis of aortic dissection and provide a basis for the rapid diagnosis of aortic dissection.However,its elevated level failed to predict the occurrence of death from aortic dissection rupture,and it can currently not be used as a biological marker to evaluate the prognosis of aortic dissection. |
