Effects Of Shengjingsan Granule Combined With Electroacupuncture On Sperm Acrosin Activity And Sperm P34H Expression In Oligoasthenospermia Patients

Objective: To investigate the effects of Shengjingsan granule combined with electroacupuncture on patients with oligoasthenospermia of kidney deficiency,and observe their effects on TCM syndrome score,sperm motility,sperm concentration,sperm acrosin activity and sperm P34 H expression,and explore the effect of Shengjingsan granule combined with electroacupuncture on sperm quality of patients with oligoasthenospermia,and preliminary discuss its mechanism of action.Methods: The study collected 105 patients with oligoasthenospermia who met the inclusion criteria.They were divided into three groups using the random number meter method: 35 cases in the electroacupuncture group,35 cases in the Shengjingsan granule group,and 35 cases in the combined group.The electroacupuncture group was treated with a Korean-style therapy device,while the Shengjingsan granule group was given Shengjingsan granule,and the combined group had both treatments performed simultaneously.One month is one treatment cycle and three cycles of treatment.Observe the TCM syndrome score,forward movement sperm(PR)ratio,sperm concentration,sperm acrosin activity and sperm P34 H expression before and after treatment in the three groups of patients,and discuss the mechanism.Results:(1)TCM syndrome scores: The TCM syndrome scores of the three groups after treatment were lower than before treatment and the difference was significant(P<0.05);after treatment,the syndrome score of the combined group was lower than that of the electroacupuncture group,there was a significant difference(P <0.05);The combination group and electroacupuncture group had no statistically significant difference compared with Shengjingsan granule group(P >0.05).(2)Sperm motility and concentration: After treatment,the PR ratio and sperm concentration of the three groups were higher than before treatment and the difference was significant(P <0.05).After treatment,the PR ratio and sperm concentration of the combined group were higher than those of the electroacupuncture group,which were significant difference(P <0.05);PR ratio of combined group was higher than Shengjingsan granule group and there was a significant difference(P <0.05),sperm concentration compared with Shengjingsan granule group,the difference was not statistically significant(P >0.05);There was no statistically significant difference in PR ratio and sperm concentration between electroacupuncture group and Shengjingsan granule group(P >0.05).(3)Sperm acrosin activity: After treatment,the activity of sperm acrosin in the three groups was higher than before treatment and the difference was significant(P <0.05);after treatment,the acrosin activity in the combined group was higher than that in the electroacupuncture group and Shengjingsan granule group,the difference was significant(P <0.05);there was no statistically significant difference between the electroacupuncture group and Shengjingsan granule group(P >0.05).In correlation analysis,before and after treatment,sperm acrosin activity was positively correlated with PR ratio and sperm concentration(P <0.05).(4)Expression of sperm P34H: After treatment,the expression of sperm P34 H in the three groups was higher than before treatment and the difference was significant(P <0.05);after treatment,the expression of P34 H in the combined group was higher than that of Shengjingsan granule group and electroacupuncture group,and there were significant differences(P <0.05);There was no statistically significant difference between the electroacupuncture group and Shengjingsan granule group(P >0.05).Conclusion: 1.Shengjingsan granule combined with electroacupuncture can alleviate the TCM syndromes of patients with deficiency of kidney and oligoasthenospermia,and provide a basis for TCM diagnosis and treatment;2.The combination of Shengjingsan granule and electroacupuncture can improve sperm motility and sperm concentration in patients with oligoasthenospermia,improve sperm acrosin activity and sperm P34 H expression,and the therapeutic effect is better than that of using electroacupuncture or Shengjingsan granule alone,providing effective for clinical patients treatment;3.Through mechanism research,it is concluded that the combination of Shengjingsan granule and electroacupuncture may improve sperm acrosin activity and sperm P34 H expression,so as to improve related semen indexes and alleviate TCM syndromes,and provide theoretical guidance for clinical treatment.
