The drug combination is a crucial treatment method for the treatment of complex diseases such as cancer and HIV.This is because on the one hand,the drug combination can reduce the dose required for the treatment of a single drug,on the other hand,it can reduce possible drug-resistant virus mutants while reducing cytotoxicity.However,due to the complexity of biological systems,it is often difficult to evaluate the therapeutic effect of combination drugs,often requiring a large number of clinical trials,which greatly increases the cost of trials.In this regard,in order to select the optimal drug combination,this requires a good trial design method,on the one hand can provide sufficient effective information for subsequent analysis,on the other hand,good design methods can often reduce the number of trials Time and reduce the cost of testing.Although many statisticians have proposed many effective methods and models,most of them are in the field of two-drug combination and few in the research of three-drug combination.Based on the Hill-based response surface model,this paper constructs a three-drug mixture design with shaft design,and uses simulated annealing algorithm for computer simulation,compares the design with uniform design and random design,and compares MSE and RSS to obtain This design is better than the uniform design republican random design.
