| Colon cancer(CRC)has become a serious disease endangering human health.Because colon cancer has different molecular phenotypes and high mortality.Therefore,it is urgent to determine the biomarkers of colon cancer.E3 ubiquitin protein ligase(HACE1)plays a role of tumor suppressor gene in many cancer processes.Histone methyltransferase LSD1 was the first histone methyltransferase found.Since histone methylation modification is reversible,more attention has been paid to LSD1.Tetraspanin 8(TSPAN8)is associated with cancer metastasis in many cancers.The purpose of this study is to explore the mechanism of HACE1,LSD1 and TSPAN8 in the metastasis of colon cancer.In this study,we found that the expression of HACE1 was low in colon cancer cells.By knocking down HACE1 and overexpressing HACE1 in colon cancer cells,we found that HACE1 could inhibit the metastasis of colon cancer,and HACE1 could inhibit the metastasis of colon cancer by inhibiting the expression of YAP1,a downstream factor of Hippo pathway.This study found that LSD1 and TSPAN8 were highly expressed in colon cancer cells.Knocking down LSD1,adding LSD1 inhibitor(Tranylcypromine hemisulfate)and knocking down TSPAN8 could inhibit the metastasis of colon cancer.This study found that LSD1 could promote the occurrence of EMT pathway downstream.That indicated that LSD1 could promote tumor metastasis by promoting the occurrence of EMT.At the same time,it was found that LSD1 regulated the expression of TSPAN8 by influencing H3k9me2 by immunoprecipitation of chromatin,which indicated that LSD1 affected the downstream EMT pathway through regulating the expression of TSPAN8 and then affected the metastasis of colon cancer.In conclusion,HACE1 can inhibit the metastasis of colon cancer by inhibiting the expression of YAP1 in Hippo pathway.LSD1 regulates the expression of TSPAN8 and affects the downstream EMT pathway,thereby affecting the metastasis of colon cancer.These results suggest that HACE1 and LSD1 are potential molecular targets for the treatment of colon cancer,which may provide new targets for the treatment of colon cancer and provide new ideas for early intervention of colon cancer metastasis. |
