Ozone is a very active gas,which is easy to decompose and has strong oxidizability.Ozone can oxidize and kill bacteria,viruses and other microorganisms.Ozone has also been shown to help stop bleeding and heal wounds.As a kind of colloidal gel with continuous phase in water,hydrogel has many medical and industrial applications,such as controlled-release and sustained-release drugs.However,so far,there are few reports on sustained release ozone hydrogel.In this study,the effects of different media and their composition on the stability of ozone were tested and analyzed,using carbomer as matrix,a kind of hydrogel with slow release of ozone was successfully prepared.The effect of sustained release ozone hydrogel on stabilizing ozone was measured by iodometry,and the microstructure of sustained release ozone hydrogel was observed by scanning electron microscope.The experimental results show that the stable ozone release time of Carbomer can be extended to 2 weeks,and the hydrogel has a dense mesh pore structure and good water absorption and moisture retention,the ozone water solution can be loaded and fixed in the grid.Then the safety of the sustained release ozone hydrogel was evaluated by mouse skin irritation test、MTT cytotoxicity test and vitro hemolysis test.The skin irritation test showed that sustained release ozone hydrogel had no irritating and allergic effect on mouse skin.MTT test showed that sustained release ozone hydrogel was low toxic to mouse fibroblast cells(L929).Vitro hemolysis test showed that sustained release ozone hydrogel will not cause erythrocyte hemolysis;The effect of the hydrogel was observed through the liver cutting bleeding model of SD rats and skin wound healing experiment.The results showed that the middle concentration gel group(containing ozone concentration 7.2±0.5ppm)and high concentration gel group(containing ozone concentration 16.4±1.2ppm)had significant hemostatic effect compared with other groups,and the hemostatic effect of high concentration group was better.The hemostatic time was about 60 seconds shorter than that of positive control group Celox~?hemostatic powder.It shows faster hemostatic effect.Sustained release ozone hydrogel in middle concentration group(containing ozone concentration 7±1.2ppm)and high concentration group(containing ozone concentration 15±3.0ppm)could significantly promote wound healing,and the early intervention effect of high concentration ozone sustained release hydrogel was better.It showed that the sustained release ozone hydrogel can improve the stability of ozone aqueous solution,delay the half-life of ozone decomposition,accelerate hemostasis and promote wound healing,and has good biosafety.It is expected to develop a new wound hemostatic material with excellent performance.
