Objective:Based on the pathogenesis theory of "deficiency of kidney and bone wilt,stagnation of qi and blood stasis" and taking "tonifying the kidney and activating blood" as the main treatment,this paper establishes a rabbit model of traumatic osteonecrosis of the femoral head in the early and middle stages to study tonifying the liver and kidney.The effects of Herba Morinda officinalis on traumatic necrosis of femoral head in rabbits in early and middle stages are studied,and the effects of reinforcing liver and kidney and strengthening bones on osteoblasts in hypoxic state are discussed in order to provide a theoretical basis for clinical application of euphorbia officinalis.Methods:The early-term traumatic femoral head necrosis model is created by using 56 adult healthy male Japanese white rabbits.52 rabbits who are successful in modeling are selected,which are divided into three groups: low dose group,middle dose group,high dose group and model control group.Each group has 13 rabbits(one in each group is a spare model rabbit).Based on the pathogenesis theory of "kidney deficiency and blood stasis,stagnation of qi and blood stasis" and "tonifying the kidney and promoting blood circulation" as the therapeutic method,the Chinese medicine Morinda officinalis is given low,middle,high dose and normal saline respectively,and I observe and detecte these quotas at the 4th and 8th week after gavage.And I look at rabbit mobility,hair,mental condition,diet.X-ray is taken to observe the shape of the femoral head,internal sclerosis,cystic degeneration and bone mineral density of the femoral head in rabbits.The blood calcium and serum alkaline phosphatase are extracted from the vein of the ear of the rabbit,and the blood oxygen partial pressure is monitored by the extraction of the rabbit’s deep arterial blood.Four rabbits are randomly selected and sacrificed to make the bone tissue sections of the femoralhead.After HE staining,the morphology of bone cells is observed under light microscope,and the rate of bone vacuole and the morphology and quantity of osteoblasts are evaluated.The data indexes of each index were observed and recorded.Finally,the data of each group were compared and analyzed statistically,so as to explore the effect of Morinda officinalis on the prevention and treatment of traumatic femoral head necrosis in the early and middle stages.Results:Under the intervention of traditional Chinese medicine Morinda officinalis,it has obvious curative effect on rabbit traumatic necrosis of femoral head in the early and middle stages.The indexes of each group are statistically analyzed and compared at different observation time points,and the overall difference was statistically significant.At the 4th and 8th week after gavage,the low,middle and high dose Chinese medicine Morinda officinalis group are better than the model control group as a whole,such as general conditions,X-ray manifestations,blood calcium and serum alkaline phosphatase concentration,arterial oxygen partial pressure,bone resorption rate of bone cells,number and morphology of osteoblasts.The therapeutic effect of the high-dose group was the best,the middle-dose group was the second,and the low-dose group was relatively poor.Conclusion:The Chinese medicine Morinda officinalis has a good effect on the treatment of traumatic necrosis of femoral head in the early and middle stages.It can increase the concentration of blood calcium and alkaline phosphatase,improve the partial pressure of blood oxygen in the necrotic femoral head,promote the blood oxygen content in the blood supply area of the femoral head,strengthen the fracture of trabecular bone and disorder of osteonecrosis of the femoral head and reduce the percentage of empty bone lacunae.The apoptosis of osteoblasts in femoral head is regulated by Morinda officinalisv to delay the development of osteonecrosis of femoral head.There is a positive correlation between the therapeutic effect and the dosage of traditional Chinese medicine.
