Objective:(1)Trigeminal neuralgia rat model was established by chronic compression of the trigeminal root entry zone,and observe the behavioral changes of rats after trigeminal nerve injury.(2)By observing the expression of Wnt protein and Wnt/β-catenin signal pathway in trigeminal root entry zone after operation.to explore whether Wnt signal pathway was activated after trigeminal nerve injury and the role of Wnt/β-catenin signaling pathway in the pathogenesis of trigeminal neuralgia,which probably provide new ideas for the pathogenesis and treatment of trigeminal neuralgia.Methods:(1)Sprague Dawley male rats were selected as experimental animals in this study,which was divided into two groups according to the method of random number table,namely TN group and Sham group.TN group: the right orbit was exposed and TREZ was compressed chronically,Sham group: only exposed the right orbit without compression injury.(2)The mechanical pain threshold in the right orofacial region of rats in TN group and Sham group was examed with Von Frey filament.(3)Immunofluorescence and Western blot were used to analyze the protein expression changes of Wnt 3a,which is representative member of Wnt protein family,andβ-catenin,GSK 3β on day of 7,14,21 and 28 day after operation in TN group and Sham group.Results:(1)The behavioral data of rats showed that the basal pain thresholds on the of3,2,and 1 before surgery remained basically the same.The pain sensation of the TN group was dull on post-operation day 1,and the pain thresholds gradually decreased on the day of 3,7 after operation and approached the basic pain threshold.The pain threshold continued to decrease from the day of 14 to 28 after operation and significantly lower than the sham group(p<0.05);Sham group rats were hyperalgesic on the day of 1 after operation,and the pain threshold was restored to the level of basal pain threshold from day 3 to 28.(2)Chronic mechanically compression of TREZ induced orofacial hyperalgesia in rats.The Immunofluorescence histochemical staining showed that in TN group the expression of Wnt 3a,β-catenin,and GSK 3β were mainly localized at the PNS of the trigeminal nerve,and the expression of β-catenin started increasing on post-operation day 14 and reached the highest on the of day 21;the expression of Wnt 3a increased at 21 days after operation in the TN group,and no significant changes were observed at day 7,14,21,and 28 after operation of GSK 3β in the TN group.Western blot results showed that compared with the Sham group,the expression of β-catenin in the TN group was increased on the day of 14 and 21 after operation,and there was no significant change in the GSK 3β in the TN group at day 7,14,21,and 28 after operation,and it was not statistically different from Sham group(p>0.05).Conclusions:(1)After chronic trigeminal root entry zone compression injury,β-catenin and Wnt 3a were mainly expressed in the PNS end of the trigeminal nerve,and increased in the TN animal model group,suggesting that the Wnt/β-catenin signal pathway may be activated.(2)GSK 3β is mainly expressed in the PNS end of trigeminal nerve,but chronic trigeminal nerve compression injury has no significant effect on the expression of GSK 3β.Whether GSK 3β is involved in the pathogenesis of trigeminal neuralgia needs further study.
