| Objective:With the continuous updating of gene detection technology,more and more research evidences indicate that LncRNA is significantly abnormal express in bladder cancer and may be used as a molecular marker in the diagnosis of bladder cancer.In this paper,the diagnostic value of LncRNAs in bladder cancer was systematically reviewed,and the diagnostic value of LncRNA UCA1 in bladder cancer was comprehensively evaluated by means of meta-analysis,which provided the statistical basis for clinic.Method:Literature from Pub Med,Embas,Cochrane library,Web of Science and other databases was searched since from database establishment to November 2019,and comprehensive retrieval was conducted according to the proposed retrieval scheme.The two investigators independently conducted literature screening based on inclusion and exclusion criteria,and the differences were resolved by consensus.Then the quality of the included literature was evaluated using the quadas-2 standard.Risk assessment charts were made with Rev Man5.3 software.After data were extracted directly or indirectly,data were merged with stata15.1 software,and calculated by the following:sensitivity,specificity,positive likelihood ratio,negative likelihood ratio,diagnostic score,diagnostic advantage ratio,the AUC of SROC curve,heterogeneity analysis and publication bias analysis.Results:According to the rigorous screening and evaluation,a total of 15 references were included to review the sensitivity,specificity and AUC of UCA1、MALAT1、PCAT-1、SPRY4-IT1、LINC00355、H19、HOTAIR、MEG3、SNHG16、ZEB1-AS1、PVT1 and PTENP1 in the diagnosis of bladder cancer.The all above LncRNAs had high diagnostic sensitivity and specificity.Among them,8 included literature were related to the study of UCA1 in the diagnosis of bladder cancer.Therefore,a meta-analysis was performed on this study.A total of 1442 patients were enrolled.The combined sensitivity was 0.84(95%CI =0.80-0.87),specificity was 0.87(95% CI=0.77-0.93),positive likelihood ratio was 6.44(95% CI =3.46-11.99),negative likelihood ratio was 0.18(95% CI =0.14-0.24),diagnostic score was 3.56(95%ci=2.70-4.42),and diagnostic odds ratio was 35.02(95%ci =14.81-82.08).The AUC of SROC curve was 0.89(95% CI =0.86-0.91).Meta-regression and subgroup analysis indicated that ethnic differences and sample types were important factors affecting sensitivity heterogeneity,while sample size was not.Deeks funnel plot analysis indicated that there was no publication bias.Conclusions:LncRNAs are differentially expressed in bladder cancer to different degrees and possess high diagnostic sensitivity and specificity,which may become a potential molecular marker for the diagnosis of bladder cancer.In addition,the diagnostic meta-analysis of the diagnostic value of UCA1 concluded that the abnormal expression of UCA1 may be a noninvasive supplement in the diagnosis of bladder cancer in the future. |
