Objective: Pancreatic cancer is one of the most difficult diseases in malignant tumors.At present,although there are many common chemotherapy and radiotherapy methods in clinic,it is urgent to study new anticancer drugs and treatment plans for pancreatic cancer to improve the therapeutic effect of pancreatic cancer.In this paper,the active component of wogonin from Scutellaria barbata was mined through network pharmacology and its effect and mechanism of inhibiting the proliferation of pancreatic cancer cells were studied.Methods: Based on the traditional Chinese herbal medicine database and the compound information database,the active components of Scutellaria barbata were fully mined,the target proteins of each component were predicted and screened by the virtual docking platform,and the important targets were determined by the compound protein disease interaction network,combined with the bioinformatics means,GO analysis and KEGG signal pathway enrichment,analysis of the potential target genes and molecular mechanism of wogonin in the treatment of pancreatic cancer;Detection of the effect of wogonin on the proliferation of BxPC-3 cells by CCK-8 cell proliferation detection kit;Detection of the effect of wogonin on the cycle and apoptosis of BxPC-3 cells by flow cytometry;Observation of the effect of wogonin on the migration and repair of BxPC-3 cells by scratch test,and transwell experiment was used to observe the effect of wogonin on the invasion of BxPC-3cells.The molecular mechanism of the inhibitory effect of wogonin on the proliferation of pancreatic cancer BxPC-3 cells was analyzed by analyzing the enrichment results of differential mRNA,target gene of differential lncrna,target gene of differential circrna source,and GO and KEGG analysis wre used for mechanism research.Key gene expression was verified by using SYBR-q PCR.Results: The results of network pharmacology showed that wogonin was an important active component of Scutellaria barbata and participated in many cell components,biological processes and molecular functions,producing functional effects from multiple perspectives;Pharmacodynamic experiments showed that wogonin could inhibit pancreatic cancer cells in G2 / M phase,promote BxPC-3 cell apoptosis,inhibit cell migration,and significantly inhibit the invasion of pancreatic cancer cells;Full conversion the results of histograms showed that wogonin could regulate the function of chromosome separation,metaphase / anaphase transformation of cell cycle,and act on the signal pathway of oocyte meiosis,cell cycle and p53 signal pathway.The target genes of different lncrna and the genes from different circrna showed that the drugs could inhibit the proliferation of cancer cells through the metabolic pathway.Wogonin reactivates p53 in BxPC-3 cells,up-regulates the apoptotic genes BAX and Beclin-1,and inhibits the anti-apoptotic gene BCL-2.Conclusion: Wogonin has the activity of suppressing the cancer cell cycle in the G2 /M phase,promoting apoptosis,inhibiting invasion and migration,thereby achieving the induction of apoptosis of pancreatic cancer cells;Wogonin can reactivate the normal function of p53 protein and promote apoptosis of BxPC-3 cells;It can regulate the network of non-coding genes to interfere with the reprogramming of the metabolic pathway of BxPC-3 cells to achieve the activity of inducing apoptosis of pancreatic cancer cells.
