Objective:to detect the expression of LAMA3 genes between the papillary thyroid cancer cell lines TPC-1,K1,BCPAP and normal thyroid cell lines Nthy-1,the thyroid papillary cancer tissues and adjacent to carcinoma tissues.Then build a cell model.A series of function test in vitro was conducted and explore the function of LAMA3 gene in the thyroid papillary carcinoma.Methods:1)collect the samples of carcinoma and para-carcinoma tissues from 56 patients with thyroid papillary carcinoma.Resuscitation and culture of thyroid papillary cancer cell lines BCPAP,TPC-1,K1 and human thyroid normal cell line Nthy-1;qRT-PCR and Western Blot techniques were used to detect the expression of LAMA3 gene at the tissue and cell levels,respectively.Synthesis and analysis of experimental results and sequencing results.2)shRNA gene knockdown technique was used to construct LAMA3 gene knockdown cell models in thyroid papillary cancer cell lines BCPAP,TPC-1 and K1.Fluorescence microscopy and qRT-PCR were used to detect the gene knockout efficiency,and the available cell models were screened out.3)the influence of lower LAMA3 gene expression on the proliferation,migration and invasion in thyroid papillary cancer cells were investigated by cck-8,cell scratch and transwell experiments.Results:1)In the papillary thyroid cancer,the mRNA experssion of LAMA3 gene was significantly high at both cell and tissue levels,the protein experssion of LAMA3 gene was significantly high at cell level and high at tissue level without significant.2)LAMA3-siRNA-1 plasmid was transformed into K1 and TPC-1 cells to construct a cell model with a significantly low expression of LAMA3 gene.3)In K1 cells,the lower expression of LAM A3 gene inhibited cell proliferation,migration and metastasis significantly.LAMA3 gene had no significant effect on the proliferation,migration and metastasis in TPC-1 cells.Conclusion:The experssion of LAMA3 gene was significantly high in thyroid papillary carcinoma and can promote the proliferation,migration and invasion of thyroid papillary carcinoma cell line K1 significantly.In this paper,the further study of LAMA3 gene may help us to find a more effective entry point for the diagnosis and treatment of thyroid papillary carcinoma.
