Objective The expression rules of NSS(Neurological Severity Scores),Wnt/β-catenin signaling pathway,BDNF(Brain Derived Neurotrophic Factor)and NGF(Nerve Growth Factor)in rats with severe cranial Brain injury at the acute stage were observed dynamically,and the intervention effect of Huoxue Huayu Decoctions on Nerve regeneration and repair was also observed.Method A total of 306 SD male rats were randomly divided into normal group,model group,brain protein hydrolysate group,Taohong Siwu Decoction group,Xuefu Zhuyu Decoction group,Tongqiao Huoxue Decoction group,Buyang Huanwu Decoction group.The severe traumatic brain Injury rat model was prepared by the modified Feeney’ free fall method except the normal group.After successfully modeling,the brain protein hydrolysate group,the brain protein hydrolysate group,Taohong Siwu Decoction group,Xuefu Zhuyu Decoction group,Tongqiao Huoxue Decoction group,Buyang Huanwu Decoction group were intragastrically given the corresponding drugs 1.11 mg/200 g,2.04 g/200 g,3.12 g/200 g,1.92 g/200 g,5.74 g/200 g respectively.The normal group and the model group were given the same volume of normal saline,and each group was intragastrically administered once a day for 7 days.Rats in each group were randomly selected for detecting NSS(neurological severity scores)on the 1st,3rd and 7th day after gavage.The contents of Wnt3 a andβ-catenin mRNA in hippocampal tissue were detected by RT-PCR.The content of BDNF and NGF in hippocampal tissue was determined by immunohistochemistry.Results(1)There was no significant change in NSS in the normal group during the experiment.The NSS in the model group was significantly increased(p<0.05),and gradually decreased with the extension of time,and reached the lowest level on the 7th day(p<0.05).Compared with the model group,the NSS of the brain protein hydrolysate group and the Taohong Siwu Decoction group decreased on the 7th day,and the NSS of the Xuefu Zhuyu Decoction group,Tongqiao Huoxue Decoction group and Buyang Huanwu Decoction group decreased on the 1st day.No significant changes were observed in the NSS on the 1st and the 3rd day(p>0.05),but the NSS decreased to the lowest level on the 7th day(p<0.05).(2)Compared with the model group,the expression of Wnt3 a mRNA in the brain protein hydrolysate group increased on the 1st day and continued to increase with time(p<0.05),but there was no significant difference between the3 rd and 7th day(p>0.05).The expression of Wnt3 a mRNA in the four decoction groups did not change significantly on the 1st day(p>0.05).The expression of Wnt3 a mRNA in the Taohong Siwu Decoction group began to increase on the3 rd day and reached a peak on the 7th day(p<0.05).The mRNA expression of Wnt3 a in Xuefu Zhuyu Decoction,Tongqiao Huoxue Decoction and Buyang Huanwu Decoction decreased on the 3rd day,and began to increase on the 7th day,which was higher than that in the brain protein hydrolysate group(p<0.05).Among them,the most significant increase in the expression of Wnt3 a mRNA was in Buyang Huanwu Decoction group,followed by Tongqiao Huoxue Decoction group.(3)Compared with the normal group,the expression of β-catenin mRNA in the model group increased significantly on the 1st day after injury,and increased with the extension of time(p<0.05).Compared with the model group,the expression of β-catenin mRNA in the brain protein hydrolysate group did not change significantly on the 1st day after injury,and increased from the 3rd day after injury,and lasted until the 7th day(p<0.05).The mRNA expression ofβ-catenin mRNA in Taohong Siwu Decoction group decreased from the 1st to3 th day,Xuefu Zhuyu Decoction group decreased on the 1st day(p<0.05),there was no significant change on the 3th day and Tongqiao Huoxue Decoction group gradually increased with the extension of time(p<0.05).There was no significant change from the 1st to 3th day in Buyang Huanwu Decoction group.The mRNA expression of β-catenin in 4 groups of Huoxue Huayu Decoction was significantly increased on the 7th day(p<0.05).Among them,the most significant increase in the mRNA expression was in Buyang Huanwu Decoction group,followed by Tongqiao Huoxue Decoction group,and the increase was greater than that in the brain protein hydrolysate group.(4)Compared with the normal group,the expression of BDNF and NGF in the model group increased significantly on the 1st day,and gradually increased with the extension of time,reached a peak on the 3rd day after injury,and then decreased on the 7th day after injury(p<0.05).Compared with the model group,the expression of BDNF and NGF did not increase until the 7th day in the brain protein hydrolysate group(p<0.05).The expressions of BDNF and NGF in Buyang Huanwu Decoction group and Tongqiao Huoxue Decoction group increased immediately on the 1st day after injury(p<0.05).The expressions of BDNF and NGF in the Taohong Siwu Decoction group and Xuefu Zhuyu Decoction group did not change significantly on the 1st day,but decreased significantly on the 3rd day(p<0.05),and then increased on the 7th day after injury(p<0.05).The expressions of BDNF and NGF were significantly increased in each Huoxue Huayu Decoction group on the 7th day after treatment(p<0.05),and the increase was significantly higher than that in the model group and the brain protein hydrolysate group.Conclusion Taohong Siwu Decoction,Xuefu Zhuyu Decoction,Buyang Huanwu Decoction and Tongqiao Huoxue Decoction have certain curative effects on the nerve regeneration and repair of rats with severe traumatic brain injury at acute stage,but the intensity of the effect is different,Buyang Huanwu Decoction and Tongqiao Huoxue Decoction have a better effect.Objective tissue were observed on the 1st,3rd and 7th day,and the contents of serum microtubule associated protein Tau and glial fibrillary acidic rotein(GFAP)were detected.Results(1)During the experiment,there was no death in the normal group and the brain protein hydrolysate group.The deaths of the other groups were mainly during the first 4 days.Compared with the normal group,the death rate of the model group,the Taohong Siwu Decoction group and the Xuefu Zhuyu Decoction was significantly increased(p<0.01).Compared with the model group,the death rate of the Taohong Siwu Decoction group and the Xuefu Zhuyu Decoction group were also significantly increased(p<0.01).There was no significant difference between the Tongqiao Huoxue Decoction group and the Buyang Huanwu Decoction group(p>0.05).(2)Compared with the normal group,the brain histopathological scores and serum Tau and GFAP levels in the rats in all treated groups were significantly increased at 3 time points(p<0.05).Compared with the model group at the same time point,the pathological scores of the rats in the Xuefu Zhuyu Decoction group,the Tongqiao Huoxue Decoction group and the Buyang Huanwu Decoction group increased on the first day,and the serum GFAP protein level of the Taohong Siwu Decoction group,the Xuefu Zhuyu Decoction group and the Buyang Huanwu Decoction group was increased(p<0.05).On the 3rd day,in all groups except the Taohong Siwu Decoction group,the pathological scores were lower than those of the model group;The serum Tau and GFAP levels of the brain protein hydrolysate group,the Tongqiao Huoxue Decoction group and the Buyang Huanwu Decoction group decreased(p<0.05).On the 7th day,the pathological scores,serum Tau and GFAP levels of all treated groups decreased(p<0.05)Conclusion In the acute stage of severe traumatic brain injury,the use of activating blood circulation and resolving blood stasis drugs will aggravate the illness and increase the risk of death,among which the Taohong Siwu Decoction and Xuefu Zhuyu Decoction have a greater risk.The best time to treat the acute phase of severe traumatic brain injury with activating blood circulation and resolving blood stasis drugs may be after the 4th day of injury,and the Tongqiao Huoxue Decoction and Buyang Huanwu Decoction have a better effect.
