Study On The Mechanism Of Momordin Ic Inhibiting The Proliferation Of HN Cancer Cells | | Posted on:2021-04-29 | Degree:Master | Type:Thesis | | Country:China | Candidate:W W Wang | Full Text:PDF | | GTID:2504306029956659 | Subject:Pharmacy | | Abstract/Summary: | PDF Full Text Request | | Objective:In order to explore the mechanism of Momordin Ic inhibiting the proliferation of head and neck cancer cells.Methods:In order to study on the mechanism of Momordin Ic inhibiting the proliferation of HN cancer through the following methods cells.(1)High-throughput drug screening system was adopted for drug screening.(2)MTT assay was used to determine whether Momordin Ic inhibited the proliferation and cell viability of head and neck cancer cells.(3)The inhibition of HN30 cell migration by Momordin Ic was detected by scratch test.(4)Annxin V-FITC/PI staining and WB were used to detect whether Momordin Ic promoted HN30 cell apoptosis.(5)Whether Momordin Ic induced MMP depolarization of HN30 was detected by JC-1 staining.(6)MTT assay,Annxin V-FITC/PI staining method and WB were used to explore the mechanism of Momordin Ic inhibiting HN30 proliferation.Results:(1)Momordin Ic screened through the high-throughput drug screening system can effectively inhibit the proliferation of head and neck cancer cells.(2)Momordin Ic can inhibit the proliferation and cell viability of head and neck cancer cells effectively by MTT assay.(3)It was determined by scratch experiment that Momordin Ic could inhibit the migration of HN30 of HNC cells effectively.(4)Annxin V-FITC/PI staining showed that Momordin Ic could inhibit the anti-apoptosis ability of HN30 in HNC cells effectively,and it was confirmed that Momordin Ic could promote the expression of pro-apoptotic protein Bax and inhibit the expression of anti-apoptotic protein Bcl-2 by detection of related proteins with WB.(5)It find that Momordin Ic induces mitochondrial membrane potential depolarization to promotes HNC cells apoptosis by JC-1 staining.(6)It was found that the Momordin Ic can inhibit the proliferation of HN30 cells by regulating the MAPK signaling pathway,PI3K/Akt signaling pathway,Wnt/β-catenin classical signaling pathway,and affecting the expression levels of c-Myc and P53.It was detected by MTT method,Annxin v-fitc/PI staining method and WB.Conclusion:Momordin Ic achieves anti-head and neck cancer activity by regulating MAPKs signaling pathway,inhibiting the activation of PI3K/Akt signaling pathway and Wnt/β-catenin classical signaling pathway mainly. | | Keywords/Search Tags: | Momordin Ic, Head and Neck cancer, HN30 cells, MAPKs, PI3K/AKT, Wnt/β-catenin | PDF Full Text Request | Related items |
