| Objective: To explore the therapeutic effect of TV2 p coating agent on MCAO/R rats and its preventive and protective effects on myocardial ischemia mice and rats.Methods: The MCAO/R model was used to investigate the therapeutic effects of different treatment time windows of TV2 p coating agents on cerebral ischemia rats.Rats were randomly divided into three large groups with different treatment treatment time windows.Each large group contained three groups of low,medium and high doses of Shuxuetong positive drug and TV2 p coating agent.These three groups shared the sham operation group and the model group.MCAO operation was performed on the experimental group,MCAO operation was performed in the experimental group.The therapeutic effect of TV2 p coating agent on MCAO/R rats was evaluated by neurological deficit score,cerebral infarction area,lactic dehydrogenase and lipid peroxide expression.The protective effect of TV2 p coating agent on myocardial ischemia was studied in mice with myocardial ischemia induced by ISO.The mice were randomly divided into 6 groups: sham operation group,model group,compound Danshen dropping pills positive drug group and the TV2 p coating agent low,medium and high dose groups,and were given administration for 6 days.From the fifth day,after 30 min administration,in the experimental group,the model of myocardial ischemia was induced by Intraperitoneal injection ISO 5 mg/kg for two days,and the mice were killed after administration for 2 hours on the 6th day.The protective effect of TV2 p coating agent on myocardial ischemia induced by ISO in mice was evaluated by detecting the expression of SOD,MDA,AST,CK and LDH in myocardial tissue and myocardial sections.The protective effect of TV2 p coating agent on myocardial ischemia was studied in rats with PIT myocardial ischemia.The rats were randomly divided into 6 groups: sham operation group,model group,compound Danshen dropping pill positive group and the low,middle and high dose groups of TV2 p coating agent,and were given administration for 6 days.After the rats were given 0.5 h on the 6th day,in the experimental group,the myocardial ischemia model was induced by sublingual intravenous injection of PIT 0.8 U/kg,and the protective effect of TV2 p coating agent on myocardial ischemia induced by PIT was evaluated by detecting the four items of blood coagulation and heart rate and blood pressure.Results: The therapeutic result of MCAO/R with different treatment time windows of TV2 p coating agent showed that it could significantly reduce the neurological deficit score and the area of cerebral infarction in rats with cerebral ischemia-reperfusion on three different treatment time window administrations.Except that there was no significant difference in the expression of LDH between the high dose group of first treatment time window and the model group,the other drug groups could significantly reduce the expression of LDH and lipid peroxide.The protective effect of TV2 p coating agent on myocardial ischemia induced by ISO in mice showed that except that the low dose group had no significant effect on the expression of AST and LDH,the other dose groups could increase the expression of SOD and decrease the expression of CK,AST,LDH and MDA in varying degrees.Compared with myocardial ischemia section,there was no large area rupture of myocardial fibers in each dose group,and only a few cells in the medium dose group had slight swelling.The protective effects of TV2 p coating agent on myocardial ischemia induced by PIT in rats showed that PT,TT and FIB expression were prolonged in different dose groups,and APTT was prolonged in high dose group;The heart rate and the maximum pressure rise rate of the left ventricle were reduced to varying degrees in each dose group.In addition,the left ventricular systolic pressure was significantly reduced in the high and medium dose groups,and the arterial systolic pressure was reduced in the high dose group.Conclusions: TV2 p film has a good therapeutic effect on cerebral ischemia in MCAO/R rats,and a better therapeutic effect can be achieved by controlling the therapeutic time window within 4 hours.The mechanism may be related to the decrease of the expression of lipid peroxide and lactic dehydrogenase.TV2 p coating agent has a good preventive and protective effect on myocardial ischemia.Its mechanism may be related to the decrease of the expression of CK,ASD and LDH,the increase of the expression of SOD,and the improvement of heart rate and blood pressure,and so on. |
