Cancer is one of the major diseases that endanger human health.Conventional surgical resection,radiotherapy and chemotherapy have their limitations.The development of nanotechnology offers new strategies for the treatment of cancer.In this paper,we gradually designed and prepared nanomaterial that can be activated by X-ray to produce reactive oxygen species(ROS),and further explored their applications in tumor theranostic after functionalization.In chapter 1,based on the current situation of cancer,we introduce the application of molecular imaging technology and nanotechnology in cancer theranostic.The synthesis,functionalization and application of metal-organic nanostructure(MON)with excellent properties in cancer treatment were discussed.A new kind of X-ray induced dynamic therapy(XDT)is described in detail,and the principles,design and its development status in tumor theranostic are introduced.In chapter 2,considering the easily excited property and unique excited-state electron transformation manner of the TiO2/Ru combination system,we firstly studied the cis-bis(isothiocyanato)bis(2,2’-bipyridyl-4,4’-dicarboxylato)ruthenium(Ⅱ)(N3)was modified on the surface of the commercially available TiO2 nanoparticles(P25),and the TiO2 nano-sheet(TiO2 NS)was prepared and also modified with the N3.This system is used as a preliminary research model to test its XDT performance.In addition,TixOy/Ru metal-organic nanostructure was further prepared,and Zr was introduced into the synthesis to stabilize the nanostructure,increase X-ray absorption and CT imaging.The energy conversion was enhanced by the unique spatial structure of MON and the TiO2/Ru composite system.Finally,after modifying Gd ions used for magnetic resonance imaging(MRI),a multifunctional and efficient nano-theranostic agent was prepared and demonstrated excellent imaging and XDT performance.In chapter 3,the biological evaluation of multifunctional and efficient XDT nanomaterial was carried out.The preparation shows low toxicity in cells and effective cancer cell killing after X-ray irradiation,and shows excellent imaging and therapeutic effects in mouse tumor models,and realized CT/MRI-guided X-ray induced cancer enhanced therapy.
