Objective This research was aim to approach the signal transductive mechanism of Ginsenoside Rg1 on myocardial hypertrophy induced by AngⅡ in neonatal rats cardiac myocytes via investigating the effect on of Ginsenoside Rg1 on myocardial hypertrophy,energy metabolism and the expression of SIRT1 and PGC-1α.Methods The primary neonatal rat cardiac myocytes were cultured,and the hyperrtophy was induced by AngⅡ(0.1 μmol/L),and the effects of different doses(12.5,25,50 μmol/L)of Ginsenoside Rg1 and SIRT1 blocker EX527(1μmol/L)on cardiomyocyte volume,total protein contents,the content of FFA,ATP and AMP,the activity of CCO in myocardial mitochondri and the protein expression of ANP,Cyt-C,SIRT1 and PGC-1α were observed.The cardiomyocyte volume was measured by computer photograph analysis system.Total protein contents were assayed by the method of Bradford.Western blot was used to quantify the protein expression of ANP,Cyt-C,SIRT1 and PGC-1α;Elisa was used to quantify adenosine triphosphate(ATP),adenosine monophosphate(AMP)and free fatty acids(FFA);The mitochondri of cardiomyocytes was saqarated by centrifugation,and the activity of CCO was assayed by Ultraviolet analytic method。Compared with the control group,the cell size,total protein content,protein expression of ANP,FFA were increased,then ATP/AMP,the activity of CCO and the expression of SIRT1 and PGC-1α was decreased in model group.Ginsenoside Rg1(25,50 μmol/L)could remarkably down-regulate the cell size,total protein content,ANP expression and FFA content,then ATP/AMP,the activity of CCO and the expression of SIRT1 and PGC-1α was increased.And the effect of Ginsenoside Rg1 was dose-relative.EX527 has no effect on the hypertrophic cadiomyocytes,and the effect of Ginsenoside Rg1(25 μmol/L)on cardio-protect and PGC-1α could be blocked by EX527.ResultsConclusions Ginsenoside Rg1 had the inhibitive effect on cardiac hypertrophy in neonatal rats cardiac myocytes induced by AngⅡ.Ginsenoside Rg1 could also ameliorate energy metabolism of myocardial hypertrophy.Ginsenoside Rg1 could up-regulate the expression of SIRT1 and PGC-1α,which indicated that the regulate effect of Ginsenoside Rg1 on hypertrophy may be via the SIRT1/PGC-1α signal pathway.The up-regulate effect of Ginsenoside Rg1 on PGC-1α could be blocked by EX527,which indicated that the regulate of PGC-1α may be via the SIRT1,then in other words,PGC-1α may be the down-regulator of SIRT1.
