ObjectiveTo investigate whether hesperetin can recover retinal damage caused by sodium iodate in vitro and in vivo and its potential mechanism.MethodsIn vitro experiments,ARPE-19 cells were divided into control group,sodium iodate(SI)group and hesperetin(HES)+SI group.The control group received normal medium,the SI group received medium containing 1200 μg/m L SI,the HES+SI group received 80 μM HES and 1200 μg/m L SI,the ARPE-19 cell apoptosis and production of reactive oxygen species(ROS)was detected in each group.Autophagy in ARPE-19 cells was detected by Ad-m Cherry-GFP-LC3 B.Extract ARPE-19 protein in the cell,western blot detection of Nrf2 / HO-1,P62 and LC3 B Ⅱ/Ⅰ related protein changes.In vivo experiments,male C57BL/6J mice were randomly divided into Control group(Control),model group(SI),and treatment group(HES+SI).After 1 week of daily intraperitoneal injection of hesperetin 50mg/kg in the HES+SI group,both the SI group and the HES+SI group were given one intravenous injection of sodium iodate(25mg/kg).The HES+SI group was given intraperitoneal injection of hesperidin(50mg/kg · d).After SI treatment,1w,2w,3w and 4w were measured by optical coherence tomography(OCT)and electroretinography(ERG)respectively.After 4w,the mice were sacrificed and their eyes were embedded in paraffin and stained with hematoxylin and eosin(HE).Thechanges of retinal structure were observed,and the apoptosis of retinal cells was detected by Td T-mediated d UTP Nick-End Labeling(TUNEL)method.ResultsHES reduced SI-induced apoptosis and intracellular ROS production in ARPE-19 cells,and significantly inhibited the occurrence and development of autophagy in ARPE-19 cells induced by SI.HES alleviated SI-induced retinal thinning and pigmentation epithelial disorders in vivo,and to some extent protected visual function.western blot showed that HES treatment increased Nrf2 / HO-1 expression,reduced P62 expression,and decreased LC3 B Ⅱ/Ⅰratio compared to SI group.ConclusionsHesperetin through anti-oxidation,inhibits autophagy and apoptosis to reduce the ARPE-19 cell damage caused by sodium iodate,and reduces the damage of retinal structure and visual function caused by sodium iodate in vivo.
