Myocardial Infarction(MI)refers to myocardial ischemic necrosis caused by severely and persistently acute ischemia in the myocardium.MI threatens human health with high morbidity and mortality.MI can be relieved by angiogenesis,which is a very complex biological process that requires multiple factors to participate.Long non-coding RNA(lncRNA)is a type of non-coding RNA with a length of more than 200 nt,which is hardly involved in protein encoding,but it can play an important regulatory role in various disease processes.Recently,there have been many reports that lncRNA affects angiogenesis by regulating HUVECs in diseases such as cancer.However,angiogenesis after MI has rarely been reported.In this study,a small pig heart infarction model was constructed,and the lncRNA related to myocardial infarction was screened by high-throughput RNA sequencing.The candidate lncRNA was transferred by bio-cloning,q RT-PCR,vector construction,cell culture,cell transfection and other biological techniques.We explored the effects of lncRNA on angiogenesis after myocardial infarction by many biotechnologies.When the candidate lncRNA were expressed in HUVECs,we used Ed U to detect cell proliferation,used FCM to detect cell apoptosis and used Matrigel to detect the tube information in vitro.The main results of this study are as follows:1.The full length of five candidate lncRNAs was successfully amplified,which were1555 nt,819 nt,1358 nt,1018 nt and 2085 nt.After q PCR,the expression levels of five lncRNAs were consistent with the RNA-seq results.2.The target genes of five candidate lncRNAs were screened according to relevant bioinformatics,namely ANGPT2,BCL2,ATG7,VEGFC and p53.The results indicated that BCL2,ATG7 and p53 may be target genes of lncRNA2,lncRNA3 and lncRNA5.ButANGPT2 and VEGFC had no target relationship with lncRNA1 and lncRNA4.3.lncRNA1,lncRNA2 and lncRNA4 could inhibit endothelial cell proliferation,lncRNA3 and lncRNA5 could promote endothelial cell proliferation.4.lncRNA1,lncRNA2 and lncRNA4 could promote endothelial cell apoptosis,lncRNA3 and lncRNA5 could inhibit endothelial cell apoptosis.5.lncRNA1,lncRNA4 and lncRNA5 could inhibit endothelial cell tube formation,lncRNA2 and lncRNA3 could promote endothelial cell tube formation.The conclusions of this study are as follows:LncRNA1 and lncRNA4 inhibit endothelial cell tube formation in vitro via inhibiting endothelial cell proliferation.Lnc RNA3 promotes endothelial cell tube formation in vitro through promoting endothelial cell proliferation.
