| Breast cancer is one of the most common malignant tumors in women.At present,because the etiology of breast cancer is not completely clear,there are still unsatisfactory side effects in breast cancer treatment,so it is urgent to find a new treatment strategy for breast cancer.More and more studies have shown that Lnc RNA plays an important role in tumorigenesis,and it could be expected to become a new molecular target for the treatment of tumors.In this study,two Lnc RNAs related to breast cancer,LINC01234 and STXBP5-AS1,were obtained through extensive literature review and database screening.For LINC01234,we found that LINC01234 was significantly overexpressed in triplenegative breast cancer cells MDA-MB-231,and LINC01234 could significantly promote cell proliferation and migration,inhibit cell apoptosis,suggesting that LINC01234 plays an important biological role in the occurrence and development of triple-negative breast cancer.Then,we used Lnc RNA online database and Dual-Luciferase reporter assay to analyze and found that LINC01234 can be used as the "sponge" of mi R-429 to accomplish its biological functions.Further experimental results showed that mi R-429 could significantly inhibit the proliferation and migration of MDA-MB-231 cells and promote cell apoptosis.Combined with the online analysis database of target genes of mi RNA and the experimental results,it was suggested that SYNJ1 and SMAD2 may be downstream target genes of mi R-429.Meanwhile,TCGA breast cancer clinical database analysis showed that the prognosis of breast cancer patients with high expression of LINC01234 was poor.Therefore,we conclude that LINC01234 may function as a ceRNA for mi R-429 to regulate the expression of SYNJ1 and SMAD2,hence serving as an oncogentic regulator of triple-negative breast cancer.For STXBP5-AS1,we found that STXBP5-AS1 was highly expressed in MCF-7 cells,and STXBP5-AS1 could significantly promote cell proliferation and migration,and inhibit cell apoptosis.Further experiment verified that STXBP5-AS1 may regulate the occurrence and development of breast cancer by involving in the m TOR and Ras signaling pathways.In conclusion,this study explored the oncogentic function of LINC01234 and STXBP5-AS1 in breast cancer cells in both molecular and cellular level,and clarified the crucial role of these two new Lnc RNAs in the occurrence and development of breast cancer,thus our finding may provide potential new targets for breast cancer diagnosis and therapy. |
