Objective: We focus on the effects of cell metabolism in Toll-like receptor2(TLR2)-driven gastric cancer cells to verify the mechanism of tumorigentic role of TLR2 in gastric cancer.Methods: We examined the glycolysis and oxidative phosphorylation levels in TLR2-driven gastric cancer cells using Seahorse analyzer;We detected ROS levels in gastric cancer cells stimulated with P3C;We determined gene and protein expression of SOD2 in TLR2-driven gastric cancer cells;We use si SOD2 to knock down SOD2 expression to verify function of SOD2 in TLR2-driven gastric cancer cells;TLR2 and SOD2 expression were examined in gastric cancer patients by immunohistochemistry.Results: We found that TLR2 promoted glycolysis and oxidative phosphorylation and inhibited ROS level via SOD2 in human gastric cancer.TLR2 increased SOD2 expression through STAT3、JNK/MAPK、NF-κB multiple signaling pathways.TLR2 and SOD2 expression were both up-regulated in gastric cancer patiens and significantly correlated with clinical advanced stage and poor survival.Conclusion: TLR2 promoted glycolysis and oxidative phosphorylation and inhibited ROS level via SOD2 through STAT3、JNK/MAPK、NF-κB multiple signaling pathways in human gastric cancer,which clarify the mechanism of up-regulated TLR2 and SOD2 expression in gastric cancer.
