| Objective:To study the chemical constituents of the ethyl acetate of Physalis alkekengi var.Franchetii and their nitric oxide inhibiting activity and quinone reductase inducing activity.Method:The compounds were isolated from P.alkekengi var.franchetii by column chromatography of silica gel,C18 reversed-phase silica gel,Sephadex LH-20,semi-preparative HPLC and other chromatographic technologies.Their structures were established by extensive analysis of their NMR spectroscopic data,X-single crystal diffraction and comparison with literature data.The inhibitory effects on inflammatory response and oxidative stress were evaluated by NO production inhibition experiment and QR inducting activity experiment.Results:Twenty-three compounds were isolated and identified from ethyl acetate of the ethanol extract of P.alkekengi var.franchetii,including nine st eroids:4,7-didehydrophysalin B(2),physalin Z(3),4,7-didehydroneophysalin B(8),phys-alin G(9),physalin L(10),physalin P(11),physalin X(13),7β-hy-dro xyphysalin L(14),physalin O(19);three terpenoids:dehydrovomifoliol(5),3β-hydroxy-5,6-epoxy-7-megastigmen-9-one(7),rel-(3E)-4-[(1R,2R,4S)-1,2,4-trihyd-rox y-2,6,6-tri-methylcyclohexyl]-3-buten-2-one(15);three phenylpropanoids:syri-ng aresinol(12),3,4-dimethoxy-5-hydroxy-cinnamyi alcohol-9-O-β-D-glucopyranosi-de(20),sachaliside 1(23);one aliphatic compounds:n-hexacosanoic acid(1);tw o alkaloids:trans-N-feruloyl-3-O-methyldopamine(16),5-hydroxy-2-pyridinemeth anol(21);one flavonoid:chrysoeriol-7-O-β-glucopyranoside(22);one aromati-c acid:vanillic acid(17);three miscellaneous types of compounds:7-epilolioli-d e(4),pubinernoid A(6),tetillapyrone(18).Compounds 2,3 and 9 show si-g nificant inhibition of NO production activity.Compound 8 shows moderate inh ibition of NO production activity.Compounds 6 and 16 can inhibit the form-a tion of NO.Compounds 2、3、5、8、9、19 show significant QR inducting a-ct ivity.Compound 16 shows moderate QR inducting activity.Conclusions:In the present research,compound 20 was new compound.Compound 5 and 17 was isolated from plants for first time.Compounds 4、6、12、15、16、18、21 and 23are reported for the first time in the genus Physalis.Compounds2、3、8、9 and 16 had significant dual inhibitory effects on inflammatory response and oxidative stress. |
