| ObjectiveTo observe the intervention effect of Qingguang an Granule on autophagy related factors ATG5 and p62 in filtering scar tissue after establishment of a rabbit model of high intraocular pressure filtration.MethodsThirty New Zealand white rabbits with chronic intraocular hypertension were randomly divided into 5 groups,6 in each group,namely blank control group,simple operation model group,positive control(+MMC)group,Qingguang’an group and negative control group(PBS)by intragastric administration.The drug was administered on the second day after the success of the model establishment and was administered continuously for two week.At the end of the experiment,the scar tissues of the surgical site were taken for examination,and the relative expression of ATG5,p62 RNA and their proteins in the scar tissues of each group were detected by q PCR and Western Blot.Results1.Effect of Qingguang’an Granule on ATG5 and p62 m RNA: The relative expression of ATG5 gene in scar tissue after two week using Qingguang’an Granule was higher than that of model group,blank group and negative control group(P<0.01),lower than that of positive control group using mitomycin(P<0.05);the relative expression of p62 gene in scar tissue after two week using Qingguang’an Granule was lower than that of model group,blank group and negative control group(P<0.01),similar to that of positive control group using mitomycin(P > 0.05).2.Effect of Qingguang’an Granule on ATG5 and p62 Proteins: The relative expression of ATG5 protein in scar tissue after two week using Qingguang’an granule was higher than that of model group,blank group and negative control group(P<0.01),but less than that of adding mitomycin group(P<0.01);the relative expression of p62 protein in scar tissue after two week using Qingguang’an granule was lower than that of model group,blank group and negative control group(P<0.01),but less than that of adding mitomycin model group(P< 0.01).ConclusionQingguang’an Granule can up-regulate the expression of ATG5 and itsprotein,down-regulate the expression of p62 gene and its protein,and promote autophagy,which may be one of the mechanisms of scar tissue reduction. |
