Preeclampsia(PE),which is specific to human pregnancy,impacts 8 million mother-infant pairs worldwide each year.This complication is defined as the new onset of hypertension(blood pressure of ≥140/90 mm Hg),proteinuria(≥0.3g/L or≥1+ by dipstick),and other signs of maternal vascular damage such as edema after 20 weeks of human pregnancy.Furthermore,Severe PE(s PE)is diagnosed based on a further elevation of blood pressure(blood pressure of ≥160/110 mm Hg)or any of the following: renal insufficiency,impaired liver function,pulmonary edema,uteroplacental dysfunction,fetal growth restriction and premature birth.In addition,decidualization is the transformation that the stromal compartment of the endometrium must undergo to accommodate pregnancy.This process influences trophoblast invasion,embryo implantation and the maintenance of a successful pregnancy.CTP synthetase is the rate-limiting enzyme in the de novo CTP biosynthesis in which a UTP is converted into CTP with the consumption of glutamine and ATP.It plays an important role in cell growth and energy metabolism.In this study,we investigated the association between CTPS expression in decidual tissues and preeclampsia.Through immunofluorescence,RT-PCR and Western Bloting,we found that the expression of CTPS in decidua tissues of s PE patients was significantly down-regulated.Through in vitro induced decidualization model,it was found that with the progress of decidualization,the m RNA level and protein level of CTPS were significantly increased,and immunofluorescence showed that the time of induced deciduation was different,and CTPS localization in cells was also different.In this process,differential expressions of LKB1,AMPK,RHEB and m TOR were observed.Glutamine analogues were added or si-RNA interfered CTPS inhibited the decidualization process,the process of cell decidualization was inhibited and LKB1,AMPK,RHEB and m TOR were significantly down-regulated,and AMPKαwas significantly down-regulated at the protein level.In summary,we believe that CTPS plays an important role in decidualization and is involved in cell energy metabolism in addition to nucleotide synthesis.Abnormal expression of CTPS in decidualization will lead to abnormal occurrence of decidualization and may lead to PE.
