| Objective:High Mobility Group Box 1(HMGB1)is a class of the High Mobility Group(HMG)family that is highly expressed in mammals and is a highly conserved protein.According to the current research,HMGB1 has promotion and inhibition in the development and treatment of tumors,and its effect mainly depends on the location in tumor cells and the influence of tumor microenvironment.However,the role of HMGB1 in laryngeal cancer and its mechanism are still unclear.Therefore,this study aimed to explore the effect of HMGB1 on the occurrence and development of laryngeal carcinoma and its mechanism.Methods:First,study the effect of HMGB1 on the occurrence and development of laryngeal carcinoma,using immunohistochemistry,the method was used to monitor the expression of HMGB1 in human laryngeal carcinoma tissues,and to score and analyze the expression of HMGB1 in lymph node metastasis,T stage,pathological stage,tumor cell differentiation and tumor classification.Related,simultaneous survival analysis,to see whether the expression of HMGB1 is related to the prognosis of patients.Then,Western blot and immunofluorescence were used to verify the expression of HMGB1 in Hep2 cells.The effects of scorch assay and soft agar colony formation on Hep2 cell proliferation,metastasis and invasion were observed.Secondly,HMGB1 in Hep2 cell lines was knocked down with shRNA and gene microarray analysis was performed to obtain differentially expressed genes.HCS proliferation assay was performed on 20 down-regulated genes with significantly differentially expressed genes and low number of reported genes,and the most significant proliferation-related positive genes were selected for further Quantitative real-time PCR verification of their relationship with HMGB1.Finally,the above results suggested that HMGB1 could affect the expression of LSM6 gene to some extent.Therefore,the method of studying the expression and effect of HMGB1 in tissues and cells was adopted to explore the effect of LSM6.Meanwhile,FACS method was used to detect the effect of LSM6 on cell apoptosis and cell cycle.Results:First,HMGB1 is highly expressed in human laryngeal carcinoma tissues and Hep2 laryngeal carcinoma cell lines.lt can promote the proliferation,invasion and metastasis of Hep2 cells.Survival analysis showed that HMGB1 had no significant correlation with the survival rate of patients with laryngeal cancer,but the expression of HMGB1 protein was significantly increased in cancer tissues with low degree of tumor cell differentiation,indicating that HMGB1 can promote the occurrence and development of laryngeal cancer.Secondly,Compared with the control group after hitting reduction with shRNA Hep2 cells have raised 149 genes,in 355 by gene,to choose the line cut 20 genes HCS proliferation detection,including LSM6 positie for proliferation related genes,compared to the other genes in this study has a stronger role in promoting proliferation,further illustrate the silence the expression of HMGB1 can reduce LSM6 gene expression,prompt HMGB1 can to some extent affect LSM6 gene expression.Thirdly,LSM6 is highly expressed in human laryngeal carcinoma tissues and Hep2 laryngeal carcinoma cell lines,and can promote the proliferation,invasion and metastasis of Hep2 cell lines,and also promote cell apoptosis,regulate cell cycle,and increase S phase and G2 phase.cell.Moreover,the expression of LSM6 was more in patients with T stage or advanced stage of clinical stage.Although the expression of LSM6 protein had no statistically significant effect on prognosis,its high expression had a tendency to shorten the life span of patients.Conclusions:Both HMGB1 and LSM6 are highly expressed in human laryngeal carcinoma tissues and Hep2 cells,and both can promote the proliferation,metastasis and invasion of Hep2 cells,which may also have adverse effects on the prognosis.HMGB1 promotes the development of laryngeal cancer may have a certain relationship with LSM6. |
