Objective:We synthesized the small interfering RNA sequence for Survivin,and the overexpression vector for Survivin,then transfected into CNE-2 cells of nasopharyngeal carcinoma(NPC).To detect the difference of Survivin expression and signal pathway PTEN/PI3K/AKT expression in nasopharyngeal carcinoma(NPC)CNE-2 cells,and to explore the effect on proliferation and apoptosis of nasopharyngeal carcinoma(NPC)cells.Methods:According to the previous studies,the sirna which has the strongest inhibitory effect on Survivin was selected to transfect CNE-2 cells to inhibit the expression of Survivin in the cells,and the overexpression plasmid was constructed to transfect CNE-2 cells to promote the expression of Survivin in the cells.The activity of CNE-2 cells at different times was detected by CCK8 assay.Survivin,PI3K,PTEN,AKT mRNA expression in CNE-2 cells was detected by RT-PCR assay.The expression of Survivin,PI3K,PTEN,AKT,p-AKT related proteins in CNE-2 cells was detected by Western blot.Results:We used CCK8 to detect cell activity.Compared with the control group,the activity of Survivin inhibited group was lower than that of blank control group,the activity of Survivin overexpression group was increased.The results of RT-PCR showed that the inhibition rate of Survivin mRNA expression and PI3K mRNA expression was(41.9±6.9)%and(43.3±7.2)%.Respectively.The inhibitory rate of AKT mRNA expression was(38.7±4.1)%(p<0.01).The overexpression rate of Survivin mRNA in Survivin overexpression group was(203.8±17.1)%(p<0.01),AKT mRNA overexpression rate was(239.0±34.2)%(p<0.01),and there was no significant difference in the relative expression of PI3K(p>0.05),PTEN(p>0.05)mRNA.There was no significant difference in the expression of PTEN,p-AKT protein between the Survivin inhibition group and the negative control group.The expression of AKT protein was significantly down-regulated,but the expression of PTEN,PI3K,p-AKT protein was significantly up-regulated in the Survivin overexpression group compared with the negative control group.Conclusion:Among the nasopharyngeal carcinoma(NPC)CNE-2 cells.When the expression of survivin was inhibited,the expression rate of AKT was decreased;When the expression of survivin was over-expressed,the expression rate of AKT was significantly increased.Survivin was positively correlated with the expression of AKT.When the expression of Survivin was inhibited in CNE-2 cells,it can effectively inhibit the proliferation of CNE-2 cells and induce the apoptosis of CNE-2 cells.When Survivin was overexpressed in CNE-2 cells,it could effectively promote the proliferation and inhibit the apoptosis of CNE-2 cells.Therefore,Survivin has a positive synergistic effect on cell proliferation and apoptosis.
