| Objective: To involvement of mitochondrial fission in blue light induced apoptosis in retinal ganglion R28 cells,which may provide some new strategies for the prevention and therapy of the retinal injury induced by blue light.Method: Retinal ganglion R28 cells were employed in this study.We investigated the blue light with the wavelength of 450 nm,illuminance of 1000LUX;Red light with the wavelength of 630 nm,illuminance of 1000 LUX.The expression of mitochondrial dynamic related protein and P-Drp1 exposed to blue light for different time periods(3h,6h,9h,12 h,18h and 24h)were detected with Western Blot.Observe the mitochondrial morphologic changes via use of laser confocal microscope through transfected plasmid Mito-Ds Red with lipofectamine 2000.Cell death was determined using the TUNEL cell apoptosis detection kit.The expression of Cleaved Caspase3,Pink1,LC3 B Ⅱ / LC3 B Ⅰ were detected with Western Blot.Mitochondrial membrane potential was determined using TMRE probes,Intracellular ROS formation was determined using DCFH-DA probes,and analysis by flow cytometry.Results: Blue light exposure an increase in the expression of Drp1 and p-Drp1,which accompanied by Mfn2 down-regulation,but there’s no statistical significance of Opa1.It displayed features of mitochondrial fission.Blue light exposure also increased the generation of reactive oxygen species(ROS),decreased the level of MMP,and increased in the expression of Pink1 、 LC3 B Ⅱ / LC3 B Ⅰ.Blue light exposure an increase in the expression of Cleaved Caspase3.Above results were inhibited by Mdivi-1,except the expression of LC3BⅡ/ LC3BⅠ.Conclusion: The damages of retinal ganglion R28 cells caused by blue light are related to mitochondrial fission.Blue light induced mitochondrial damage and mitochondrial dysfunction in retinal ganglion R28 cells through mitochondrial fission,which stimulated apoptosis and mitochondrial autophagy.Inhibited of Drp1,the mitochondrial fission can be inhibited,mitochondrial function will be reversed,and cells survival will be improved.This mays provide some new strategies for the prevention and therapy of the retinal injury was exposured to blue light. |
