| Objective:Codonopsis lanceolata and Polygonatum sibiricum are plants grown in Changbai Mountain and can be used as food or medicine.Codonopsis lanceolata can strengthen body strength and other functions,while Polygonatum has the functions of regulating immunity and so on.In this study,compound polysaccharides from Codonopsis lanceolata and Polygonatum sibiricum polysaccharides were used to explore their immune activity.Method:①This experiment took Codonopsis lanceolata and Polygonatun sibiricum as the experimental object,using hot water leaching method,through alcohol precipitation,removing protein,acetone,ether washing and precipitation,extraction of Codonopsis lanceolata coarse polysaccharide,crude polysaccharide of Polygonatum sibiricum.The content of total sugar in the polysaccharide was measured by the phenol sulfuric acid method.②Codonopsis lanceolata polysaccharide,Polygonatum sibiricum polysaccharide is acted on RAW 264.7 in vitro,and detected the proliferation reaction,the secretion of cytokine TNF-α,IL-6,NO,and the expression of iNOS mRNA.Co-cultured with concanavalin(ConA)stimulated spleen lymphocytes of mouse to produce IL-2 and IFN-γ.③Codonopsis lanceolata and Polygonatum sibiricum Crude polysaccharide composition were acted on RAW 264.7,and the proliferation reaction,the secretion of cytokine TNF-α,IL-6,NO concentration,and co-cultured with ConA stimulated spleen lymphocytes of mouse to produce IL-2 and IFN-y.④ The crude polysaccharide composition was gavaged into the mice according to the high dose group(800 mg/mL)and the low dose group(200 mg/mL),and the mice were given 1 times a day for 30 days.The thymus index and spleen index of mice were measured.Mouse spleen cells were co-cultured with LPS and ConA,and the number of splenic lymphocytes was detected by CCK-8.The transformation of B and T lymphocytes in mice was examined.LDH assay was used to detect the activity of NK cells when mouse spleen cells were incubated with YAC-1 cells.Result:①The experimental results showed that the total sugar content of the extracted polysaccharides was 37.25%and 13.5%respectively.②The concentration of 125~1000 polysaccharide mg/mL,TNF-α,IL-6,NO,IL-2 and IFN-yincreased with the dose increasing in dosages of Codonopsis lanceolata polysaccharides.The concentration of TNF-α and IL-6 was dose-dependent in the dose range of 111~1000 mg/mL.Within the range of 125~1000 mg/mL,NO,IL-2 and IFN-y also increased with the dosage,and the mRNA generation could be significantly increased compared with the normal group.It can be speculated that the mechanism of two polysaccharides promoting NO secretion may be the result of increasing iNOS mRNA production.③There was no significant difference in the proliferation of RAW 264.7 cells and the production of TNF-α,IL-6,NO,IL-2,IFN-γ in Codonopsis lanceolata and Polygonatum sibiricum Crude polysaccharide composition,or significantly higher than that of Codonopsis lanceolata polysaccharides or Polygonatum sibiricum polysaccharides.④In the experimental group,thymus index,spleen index,B lymphocyte transformation and NK cell activity in the high-dose group and low dose group were significantly higher than those in the normal group.The results of T lymphocyte transformation test showed that there was no significant difference between the low dose group and the normal group.The proliferation of T lymphocyte in the high dose group was significantly higher than that in the normal group.Conclusion:①Codonopsis lanceolata polysaccharides,Polygonatum sibiricum polysaccharides can promote the immunization activity of monocyte macrophage and T cell better.②Codonopsis lanceolata polysaccharides can synergy with Polygonatum sibiricum polysaccharides,so as to better exert its immune activity.It can promote the immune activity,and higher than or equal to the effect of the polysaccharide of Codonopsis lanceolata and Polygonatum sibiricum,which can better promote the immune activity of B cells and T cells.③Codonopsis lanceolata and Polygonatum sibiricum Crude polysaccharide composition can act on the body and improve the activity of B,T cells and NK cells in order to improve the non specific immunity and specific immunity effect of the body. |
