Objective: Through the establishment of collagen-induced arthritis(CIA)mice model,to investigate the anti-inflammatory and bone erosion mechanism of Periploca forrestii saponins(PFS)on the collagen-induced arthritis mice model.Methods: Collagen-induced arthritis of BLAB/c female mice induced by collagen was randomly divided into three groups: normal group,CIA model group and Periploca forrestii saponins(PFS)group.The animals were tested for each group of animals: an ankle joint volume was measured every 4 days after immunization and before immunization;Take a picture of ankle,take a symptom score;Take the ankle joint for section,observe the pathological changes;The expression of p-p65 and MMP-9 protein in synovial tissue was detected by immunohistochemistry;The expression of the related protein was detected by Western blotting;The content of CII-related antibody in the serum and the content of MCP-1 in the tissue were measured by ELISA.Results:Measurement of ankle and arthritis symptoms showed that PFS could relieve the swelling degree of the ankle joint and reduce the CIA model of ankle joint symptom score(F=67.181,P<0.05).The results of HE staining and pathology showed that PFS could inhibit synovial hyperplasia,inflammatory cell infiltration in CIA model(F=424.427,P<0.001).The results of toluidine blue staining and pathology showed that PFS could inhibit the destruction of ankle cartilage in CIA model(F=211.861,P<0.001).The results of TRAP staining and counting showed that PFS could inhibit the infiltration of osteoclasts and prevent the damage of ankle cartilage and bone(F=12.041,P<0.05).The results of immunohistochemistry showed that PFS could inhibit the expression of p-p65 and MMP-9 positive cells in CIA model(P=129.716,38.183,P<0.001).Western blotting showed that PFS inhibits protein IκB(F=83.728,P<0.01),p-IκB(F=44.737,P<0.01),p-p65,MMP-9(F=8.794,P<0.05),c-fos(F=93.899,P<0.01)and cathepsin K expression.The results of ELISA showed that PFS could decrease the expression of Ig G and Ig G2a(P<0.05)in serum of CIA model,and increase the expression of Ig G1(P<0.01)and Ig M;PFS could decrease the expression of MCP-1 in CIA model(F=83.328,P<0.05).Conclusions: PFS could inhibit the degree of joint swelling and the damage of bone in CIA model.The possible mechanism was that PFS blocked the NF-κB signal pathway,which expressed IκB,p-IκB,p-p65,MMP-9,c-fos,cathepsin K,MCP-1 levels and the expression of related antibodies.
