| Scutellaria and Rhizoma herb-pair is recorded in the Membrane-Source-Opening Beverage of Pestilential Theory.The compatibility of the two medicine can be used for early pestilence,syndrome of pathogen hidden in interpleuro-diaphramatic space and loathing of cold with high fever.The addition and subtraction prescription of the compatibility are clinically used for antipyretic and dispelling summer-heat.At the initial stage of our research,we found that the compatibility of Scutellaria and Rhizoma can significantly relieve hyperthermia in rats.This paper analyzes the chemical constitutes,metabolism in vivo and tissue distribution including heart,liver,kidneys,spleen and lung of the compounds of Scutellaria and Rhizoma herb-pair and other medicines in this prescription using the combination of in vitro and in vivo components analysis by HPLC-QTOF-MS/MS.The main achievements were obtained as follows:1.Composition analysis of aqueous extracts of single medicineHPLC-QTOF-MS/MS was used to systematically analyze the chemical constitutes of aqueous extracts of single medicine.A total of 37 compounds were isolated and identified from the aqueous extracts of Scutellaria,of which are 30 flavonoids or flavonols like Baicalin,Baicalein,Wogonin,and Oroxylin A,etal.4 flavanones or flavanols like Dihydrobaicalein,Eriodictyol,and 3 simple phenylpropanoids like 2-(3-Hydroxy-4-methoxyphenyl)-ethyl-l-O-α-L-rhamnosyl-(1→3)-β-D-(4-feruloyl)-glucoside,Martynoside and Acteaside.28 compounds were isolated and identified from the aqueous extracts of Rhizoma anemarrhenae,of which are 21 steroidal saponins like timosaponin B Ⅱ,timosaponin B Ⅲ and timosaponin D,etal.4 xanthones like mangiferin,neomangiferin and isomangiferin,2 norlignans like(z)-hinokiresinol and Hinokiresinol and 2,4’,6-trihydroxy-4’-methoxybenzophenone.2.Chemical constitutes analysis of aqueous extracts of Scutellaria and Rhizoma herb-pair.49 compounds were detected from aqueous extracts of Scutellaria and Rhizoma herb-pair,among them 33 compounds were from Scutellaria and 16 were from Rhizoma.The compounds from Scutellaria inchuding Baicalein,Norwogonin and Wogonin et 26 flavones or flavonols,4 flavanones or flavanonols like Eriodictyol,5,7-dihydroxy-6-methoxyflavanone,Dihydrobaicalein and 3,5,7,2’,6’-pentahydroxy fl avan on e,3 pherylpropanoids like 2-(3-Hydroxy-4-methoxyphenyl)-ethyl-1-O-α-L-rhamnosyl-(1→3)-β-D-(4-feruloyl)-glucoside,Martynoside and Acteaside;the compounds from Rhizoma like Timosaponin B Ⅱ,Timosaponin B Ⅲ,Timosaponin A Ⅲ et 13 steroidal saponins;Mangiferin,Neomangiferin and Isomangiferin 3 xanthones.3 In vivo metabolism study of aqueous extracts of Scutellaria baicalensis and Rhizoma herb-pair.11 prototype components and 19 metabolites were detected in plasma.6 compounds were from Scutellaria and 5 from Rhizoma.19 metabolites in plasma were monomethyl mangiferin(M1),monodehydroxyl norathyriol glucuronide(M3),baicalein 5-O-glucuronide-1-O-glucuronide(M5),baicalein 6-O-glucuronide-7-O-glucuronide(M6),Hispidulin-5-O-glucuronide-7-O-glucuronide(M27).15 prototype components and 27 metabolites were detected in urine,among them 10 compounds were from Scutellaria and 5 were from Rhizoma.27 metabolites in urine were tnorathyriol glucuronide(M1),monodehydroxyl norathyriol glucuronide(M3),baicalein 6-O-glucuronide(M7),baicalein 5-O-glucuronide(M8),Hispidulin-5-O-glucuronide-7-O-glucuronide(M27)etal.The prototype components detected in heart were Baicalin and Mangiferin;the prototype component in liver and kidneys was Mangiferin only;in spleen were Mangiferin,Baicalin,Wogonin-5-β-D-glucoside and Chrysin-7-O-β-glucuronopyranoside and in lung were Mangiferin,Baicalin and Wogonin-5-β-D-glucoside.Ten metabolites detected in heart were baicalein 5-O-glucuronide-7-O-glucuronide(M5),baicalein 6-O-glucuronide-7-O-glucuronide(M6),baicalein 6-O-glucuronide(M7)etal;six in liver were baicalein 6-O-glucuronide-7-O-glucuronide(M6),baicalein 6-O-glucuronide(M7),baicalein-6-O-glucose-7-O-glucuronide(M10)etal;that in kidneys were baicalein 6-O-glucuronide-7-O-glucuronide(M6),baicalein-6-O-glucose-7-O-glucuronide(M10),wogonin 5-O-glucuronide-7-O-glucuronide or oroxylin A 6-O-glucuronide-7-O-glucuronide(M22);ten in spleen were baicalein 5-O-glucuronide-7-O-glucuronide(M5),baicalein 6-O-glucuronide-7-O-glucuronide(M6),baicalein 6-O-glucuronide(M7),Hispidulin-5-O-glucuronide-7-O-glucuronide(M27)and 15 in lung were monodehydroxyl norathyriol glucuronide(M3),monodehydroxyl norathyriol glucuronide(M4),baicalein 5-O-glucuronide-7-O-glucuronide(M5).These results lay foundations for further research of medicinal effective components,metabolic pathways and metabolic mechanism of Scutellaria and Rhizoma herb-pair. |
