Abstract/Summary:
Saliva Plebeia is one of clinical prescription of Nanjing Drum Tower Hospital,which is effective in treating urinary tract infection.The prescription is composed of four herbs,S.plebeia,P.asiatica,V.yedoensis and S.serissoides.This paper is mainly for two aspects of research.First,The chemical of Serissa japonica was studied,which can offer chemical basis for Saliva Plebeia.Second,the anti-inflammatory mechanism was studied in order to charify the signaling pathway of the prescription.1 The chemical constituents and bacteriostatic effect of Serissa japonicaThe constituents were isolated by various chromatographic techniques(silica gel,RP-MPLC,and PHPLC)and their structures were determined on the basis of their spectroscopic data,as well as literatures.eleven compounds were obtained and identified as 3-hydroxy-1,2-dimethoxy anthraquinone(1),1,2,4-trimethoxy-3-hydroxy-6-methyl anthraquinone(2),1,3,8-trihydroxy-6-methyl-anthraquinone(3),pinoresinol(4),medioresinol(5),(7S,8R)-balanophonin-4-O-β-D-glucopyranoside(6),lupeol(7),IcarisideF2(8),gallic acid(9),methyl caffeate(10),and 4-hydroxy-3-methoxyphenyl-β-D-glucopyranoside(11).compounds 4-5 were isolated from this plant for the first time,compounds 1~3、6~11 were isolated from serissa genus for the first time.The eleven compounds were conducted the bacteriostatic activity research.The results showed that nine compounds have bacteriostatic action.2 The anti-inflammatory mechanism of Saliva PlebeiaThe use of LPS-stimulated murine macrophages RAW264.7 established in vitro model of inflammation,By using ELISA kit to detect the contents of cell inflammatory factor PGE2 in the cells secrete,by using Western blot to detect the contents of intracellular COX-2.The results showed that prescriptions can significantly reduce the contents of PGE2 and COX-2,suggesting that Saliva Plebeia can play its anti-inflammatory effects through inhibition of cyclooxygenase activation of COX-2.
