| Objective:To study the mechanism of Jurkat cells apoptosis induced by Aggregatibacter actinomycetemcomitans(A.actinomycetemcomitans)cytolethal distending toxin(CDT).Methods:After human Jurkat cells were treated by A.actinomycetemcomitans CDT for 24 hours,Real-time fluorescence quantitative reverse transcription PCR and immunofluorescence were used to examine the expression of related signaling molecules gene(ubc9/p53/dr5/caspase-8/caspase-3).Then overexpression and siRNA technology were used to process Jurkat cells.The cell apoptosis rates of Jurkat cells were examined and analyzed by FCM.Real-time fluorescence quantitative reverse transcription PCR and Western blot were used to test related signaling molecules of the pathway downstream.Results:Real-time fluorescence quantitative reverse transcription PCR and immu-nofluorescence suggested that the expression of related signaling molecules(Ubc9/P53/DR5/Caspase-8/Caspase-3)were all up-regulated.After overexpression of ubc9/p53/dr5/caspase-8 in Jurkat cells,up-regulation of related signaling mole-cules of the pathway downstream and higher apoptosis rates were observed compared to the negative control groups.While down-regulation of related signaling molecules of the pathway downstream and lower apoptosis rates were observed compared to the negative control groups after siRNA processing.Conclusion:One possible signaling pathway(Ubc9/P53/DR5/Caspase-8/Caspase-3)during Jurkat cells apoptosis induced by A.actinomycetemcomitans CDT was successfully proved,laying foundations for further understanding of the mechanism of localized aggressive periodontitis(LAgP)induced by A.actinomycetemcomitans CDT and also contributing to diagnosis and treatement of LAgP. |
