Objective Explore the influence of Capparis spinosa L.extract on the amount of urine protein,the Treg cells and Th17 cells balance,the levels of anti-ds-DNA,ANA in the serum,and renal pathology in systemic lupus erythematosus(SLE)model called MRL/lpr mice.And then discuss the treatment of Capparis spinosa L.on systemic lupus erythematosus and lupus nephritis.Methods1.24 MRL/lpr mice model and 6 normal C57BL/6 mice were randomly divided into Normal group(Normal control,N)(N=6),Model group(Model group,M)(N=6),High dose group(1.5g/kg/d)(N=6),Middle dose group,M)(1g/kg/d)(N-6),Low dose group(0.5g/kg/d)(N=6);Each intervention group was treated with Capparis spinosa L.for 4 weeks,normal group and MRL/lpr model group with the same amount of saline lavage.2.Using automatic biochemical analyzer test and calculate the quality of 24 h urine protein.3.using flow cytometry instrument test mice spleen CD4+IL-17+T cells(Th17 cells)and CD4+CD25+Foxp3+T cells(Treg cells),and caculate th17/Treg cells proportion.4.Enzyme linked immunosorbent assay(ELISA)was used to detect the concentration of anti ds-DNA antibody and anti nuclear antibody in serum.5.PE、PAS、PAM staining was used to observe the renal pathological changs.Results All 24 MRL/LPR mice and all 6 C57BL/6 mice survived until the end of the experiment,all mice are completed the test of urine protein,flow cytometry,renal pathology.1.the urine protein quantitation of 24 hours changes before and after treatment Urine protein quantitation of model group after treatment was higher than the former significantly(P<0.05).Compared with the former,urine protein quantitation of high dose group decreased significantly(P<0.05),and decreased obviously than model group(P<0.01).The levels of proteinuria in the middle dose group were slightly higher than before treatment,but lower than those in the model group(P<0.01).2.using flow cytometry instrument test treg/th17 cells of the mouse spleen cell suspension Compared with model group,th17 cells in normal control group inceased significantly(P<0.01),but treg/th17 cells ratio decreased(P<0.01).Compared with model group,treg/th17 cells in middle and high dose group increased(P>0.05).3.serum anti-dsDNA antibody and antinuclear antibodies content Compared with the normal group,serum anti-dsDNA antibody and antinuclear antibodies content in model group had increased significantly(P<0.05).Serum antinuclear antibodies in high dose group had statistical significance(P<0.05).4.Renal pathology show In middle and high dose group mice AI scores were significantly lower than that of model group(P<0.01),which was also significantly lower than the low dose group(P<0.05).High,middle,low dose group CI scores were significantly lower than model group(P<0.05).Conclusion ①Capparis spinosa L.can reduce urine protein of SLE model mice,and dose dependence was observed in the dose range.②Capparis spinosa L.can adjust the balance of treg cells and th17 cells in MRL/lpr model mice.③Capparis spinosa L.can reduce lupus mice peripheral blood concentrations of ANA.④Capparis spinosa can significantly reduce kidney damage in a mouse model of MRL/lpr.
