Objective To screen the differentiated gene expression of Scnnl a in the hepato-cellular carcinoma tissue,and established Scnnla over-express Hepa1-6 mice liver cancer cells model.Methods m RNA expression of Scnnla in cells(Hepa1-6,HepG2、Hep3B,Be17404,Huh7,Lo2)were detected by real-time q PCR.The gene expression of Scnn1α in both liver tumor tissue and adjacent tissue in patients with primary liver cancer were meas-ured by Immunohistochemistry.Briefly,Scnnla full-length c DNA sequence was cloned into the vector pcDNA3.1,named Scnn1α-pcDNA3.1,which was comfirned by DNA sequencing.Scnn1α-pcDNA3.1 was transfected into hepatocellular carcino-ma cell Hepa1-6 by liposome 2000.MTT assay,Colony-forming assay and Wound scrstch assay were performed to investigate the cell growth activity and proliferation capacity,Flow cytometry was used to examine cell cycle distribution and apoptosis rate.To establish the in vivo xenograft model,and evaluate the in vivo anti-tumor efficacy of Scnn1αsingle treatment in these liver cancer xenograft models.Results The protein expression of Scnnla was elevated in the liver cancer tissue,compare to the adjacent tissue.The combined results of Scnnla over-expression and interfering study in Hepa1-6 demonstrated that Scnnla over-expression was capable of markedly inhibiting Hepa1-6 proliferation by blocking most of cells at Gl/Sphase.Scnnla treatment could effectively inhibit the in vitro hepatocellular carcinoma cell growth.Conclusion Scnn1a expression inhibits tumor growth.
