| Object To develop a kind of anti-seawater alginate fiber wound dressing,and offer it to the wounded people who work on the sea.Methods According absorption rate to study on the hygroscopicity alginate fiber wound dressing.We used a blood-clotting test based on alginate fiber wound dressing and gelfoam.New Zealand rabbits were used for animal experiment.They were divided into 3 groups,which were gelfoam group,alginate fiber wound dressing group and gauze group.The rabbit ear artery model and rabbit full-thickness skin wounds model were established to estimate hemostyptic property.We used waterproof test,bacterial barrier test and waterproof test in movement condition to select the appropriate material as our waterproof material.The rats full-thickness skin wounds model was established to estimate wound healing effect.And then at 5th,8th,10th,randomly 3 rats were killed,and take on both sides of the spine of wound tissue,H&E staining and Masson staining were used to observe the structure of healed tissues.The expression of TGF-β and CD31 in wound tissue was observed by immunohistochemical method.Cytotoxicity assay,hemolysis test,irritation test,sensitization test and shon-term stemic toxicity test were used to study the toxicity of the alginate fiber wound dressing.Results We found that water abortion was not significantly different in alginate fiber wound dressing and gelfoam.Compared with gauze group,the water abortion of gelfoam was different(P<0.05),alginate fiber wound dressing markedly different(P<0.01).The result showed that the BCI of alginate fiber wound dressing was better than absorbable gelfoam(P<0.05).Compared with gelatin sponge,the bleeding time and the Hb absorbance values of alginate fiber wound dressing all declined(P<0.01,P<0.05).And the effect was much better compared with gauze(P<0.01,P<0.01).In the rabbit ear artery model,compared with other groups,the bleeding volume is different(P<0.05)and the clotting time is markedly different(P<0.01).Through waterproof test,bacterial barrier test and waterproof test in movement condition,we considered that sterile dressing is appropriate material as our waterproof material.In the rats full-thickness skin wounds model,on the 5th day,the wound healing rate of alginate fiber wound dressing group is markedly higher than gauze group(P<0.01),and higher than chitosan group(P<0.05).H&E staining showed that alginate fiber wound dressing could promote epidermis formation.Masson staining showed that alginate fiber wound dressing could increase the normalization of collagen distribution.In the alginate fiber wound dressing group,the expression of TGF-β was markedly higher than gauze group(P<0.01).In the alginate fiber wound dressing group,the expression of TGF-β was lower than gauze group(P<0.05)on the 10th day.In the alginate fiber wound dressing group,the expression of CD31 was higher than gauze group(P<0.05)on the 5th day.The expressionof CD31 were not different in all groups on the 8th day and the 10th day.The expression of CD31 were not different between alginate fiber wound dressing group and chitosan group(P>0.05).Meanwhile the toxicology tests are all negative.Conclusion According to above research,alginate fiber dressing has fine hemostasis property,is good for wound healing and good biocompatibility.Sterile dressing with alginate fiber dressing can be used for anti-seawater. |
