Objective To observe the effect of Ziyinyikangtiaochong decoction on ovarian weight,capillary permeability,the content of E2,inhibin B(INHB)and cystic fibrosis transmembrane conductance regulator(CFTR)in model rats of ovarian hyperstimulation syndrome so as to explore efficacy and mechanism of the decoction.Method Sixty Wistar rats of 20-day-old were randomly divided into normal group,model group,Ziyinyikangtiaochong decoction group and indomethacin group with 15 rats each group.Except for normal group,the other three groups were inclined to use pregnant mare serum gonadotropin(PMSG)and human chorionic gonadotropin(hCG)to set up the ovarian hyperstimulation syndrome model after 2 days adaptive feeding.The normal group were injected with saline 0.1 ml/day from age 22d~26d,and the other three groups were injected with PMSG 10IU from age 22d~25d,then injected with hCG 100IU on 26-day-old.One hour before using hCG,lavaged with saline,Ziyinyikangtiaochong decoction or indomethacin of each group for 7 consecutive days.On 33-day-old,rats in all groups were weighed,then anesthetized and injected with 0.1ml methylene blue dye solution via the tail vein,collect cardiac blood after 30 minutes,then put death to all of the rats,collect peritoneal liquid,and weigh the ovaries,make ovarian tissue into pathological section observed by the light microscope,detect the absorbance of methylene blue dye solution with spectrophotometer,radio-immunity assay was used to detect content of E2,enzyme-linked immunosorbent assay was used to detect content of INHB,and immunohistochemistry and western-blotting were used to detect the expression of CFTR in ovarian tissue of the rats.Results 1.Successfully established OHSS model rats Compared with the normal group,the ovarian weight significantly increased in the other three groups(P<0.01),the methylene blue dye solution content of peritoneal liquid was higher than normal group(P<0.01),vascular permeability increased,OHSS model was successfully made.2.The effects on ovarian weight and ovarian morphologic changes The ovarian weight of model group,Ziyinyikangtiaochong decoction group and indomethacin group increased significantly when compared with normal group(P<0.01),while there was no statistically significant difference between Ziyinyikangtiaochong decoction group and indomethacin group(P=0.887).From the light microscope,we can see each developmental stages of ovarian follicles in normal group rats.In model group,the rats were swelling in ovarian tissues,increasing in ovarian volume,only some developmental stages of ovarian follicles in the ovarian cortex,and the granular cell arranged irregularly,multiple follicular cysts and corpus luteum cysts,luteinizing distinctly with interstitial edema.Compared with model group,Ziyinyikangtiaochong decoction group and indomethacin group’s ovarian morphology,the number of follicular cysts and corpus luteum cysts all had a decrease.3.Detect the methylene blue dye solution content of peritoneal liquid The methylene blue dye solution content of peritoneal fluid in ziyinyikangtiaochong decoction group and indomethacin group were significantly lower than that in model group(P<0.01),and ziyinyikangtiaochong decoction group significantly lower than indomethacin group(P<0.05).4.Detect the serum E2,INHB Serum E2 and INHB level in normal group were lower than other three groups(P<0.01),in ziyinyikangtiaochong decoction group and indomethacin group were lower than that of model group(P<0.01);And the efficacy of the ziyinyikangtiaochong decoction group for lowering INHB level is obviously superior to that of indomethacin group(P<0.05).5.Detect the expression of CFTR protein in ovarian tissue Ovary CFTR protein expression of rats in the normal group was significantly lower than that in model group,indomethacin group,ziyinyikangtiaochong decoction group,with significant difference(P<0.01).The effect of Ziyinyikangtiaochong decoction,which is similar to that of indomethacin,could reduce the expression of CFTR in ovarian tissue significantly(P<0.01),in addition,slightly lower than that in indomethacin group,however with no statistically significant difference(P=0.93).Conclusion 1.PMSG joint hCG can establish the model rats of OHSS successfully.2.Ziyinyikangtiaochong decoction can reduce the ovarian weight of model rats of OHSS,and improve the ovarian morphologic changes.3.Ziyinyikangtiaochong decoction can effectively lowering serum E2 and INHB level.4.iyinyikangtiaochong decoction can reduce the expression of CFTR protein in ovarian tissue.5.Ziyinyikangtiaochong decoction can treat ovarian hyperstimulation syndrome through reducing the content of E2,INHB and the expression of CFTR in ovarian tissue and improving abdominal capillary permeability.
