Objective According to the characteristics of Oral Drug Delivery System,improve the conditions of extraction process of Cassia twig and Licorice,increase the extraction rate of total organic acids from Cassia twig and Total flavonoids from Licorice,firewood of total organic acids from Cassia twig and Total flavonoids from Licorice medicine prepared Active pharmaceutical ingredients(API).In order to provide a new Theoretical basis of Administration for the treatment of total organic acids from Cassia twig and Total flavonoids from Licorice cure Bradyarrhythmias.Methods 1.With Cinnamic acid and Cinnamic aldehyde 2 organic acids organic acids as standard,using HPLC to establish a method for determination of 2 organic acids organic acids in Cassia twig to total organic acids.Regarding the content of total organic acids、Cinnamic acid and Cinnamic aldehyde as index to screening processing methods of cassia twig.2.With Cinnamic acid as the standard,the determination method of the content of total flavonoids of Licorice established by UV.With the total flavonoids extraction rate as the evaluation index,based on single factor test,orthogonal design is used to optimize the extracting process of ethanol decoction.3.With Rutin as the standard,the determination method of the content of total organic acids of Cassia twig established by UV.With the total organic acids extraction rate as the evaluation index,based on single factor test,orthogonal design is used to optimize the extracting process of ethanol decoction.4.Using the content of the total organic acid extract of cassia twig and total flavonoids of licorice as index,Factors influencing stability of the total organic acid extract of cassia twig and total flavonoids from licorice are evaluated by high temperature test,high humidity test,strong light test.5.To establish chronic arrhythmia model of SD rat by injecting verapamil,the effect of bradycardia resistant in the total organic acid of cassia twig and total flavonoids of licorice is evaluated by rat heart rhythm,PR interval period,mortality rates.Results 1.To establish the UV quantitative analysis method of total organic acids in Cassia twig,the detection wavelength at 268nm,within the range of 1~7μg·mL-1 exhibited good linearity.Precision,stability,repeatability,recovery rate are in line with the requirements.To establish HPLC content analysis method of Cinnamic acid and Cinnamic aldehyde.There was a good linear relationship between the them respectively in range of 0.0025-0.0500μg、0.0115-0.2294μg.And precision,stability,repeatability,recovery rate test all meet the requirements.The processing method of Cassia twig determined as:honey processing,and the honey is second degree.2.The extraction technology of total organic acids in Cassia twig conditions determined as:with 10 times amount of 90%ethanol,direct fire reflux extraction 3 times,each time 1h.The preparation technology of Coptis extract,three verification test total organic acids extraction rate was 69.67%,the rate of total organic acids extract 16.40%,purity of 8.83%.3.To establish the UV quantitative analysis method of total flavonoids in licorice,the detection wavelength at 410nm,within the range of 0.0025 mg·mL-1-0.03 mg·mL-1 exhibited good linearity.Precision,stability,repeatability,recovery rate are in line with the requirements.The extraction technology of total flavonoids in licorice conditions determined as:with 35 times amount of 70%ethanol,direct fire reflux extraction 3 times,each time 1h.The preparation technology of Coptis extract,three verification test total organic acids extraction rate was 127.77%,the rate of total organic acids extract 40.85%.4.To establish the content determination of alkaloids in Rhizoma Coptidis,the content of alkaloid≥29%,by UV.The linearity,precision,stability,repeatability,recovery rate are in line with the requirements.The properties of total alkaloids in Rhizoma Coptidis is stability at high temperature of 60 ℃,75%high humidity and high light conditions.To establish the content determination of the total organic acid extract of cassia twig,the total organic acid of cassia twig and total flavonoids of licorice in extraction of licorice by UV.The linearity,precision,stability,repeatability,recovery rate are in line with the requirements.The quality of the total organic acid extract of cassia twig and total flavonoids of licorice is controled:the content of the total organic acid extract of cassia twig and total flavonoids of licorice≥29%.The properties of the total organic acid extract of cassia twig and total flavonoids of licorice is stability at high temperature of 60℃,75%high humidity and high light conditions.5.Therapeutic effect of the total organic acid of cassia twig、total flavonoids of licorice and Guizhi Gancao Decoction is evaluated by chronic arrhythmia model of SD rat injected Verapamil,the result shows that the high dose group of Guizhi Gancao Decoction has best effect on chronic arrhythmia.Conclusion Through single factor and orthogonal experiment to optimize the extraction and purification process of total organic acids in Cassia twig and total flavonoids in licorice extract conditions,obtained the content is above 60%.And use UV and HPLC to control the quality of the component index of total organic acids in Cassia twig and total flavonoids.Secondly,...
