| Objective Analysing the different time periods’ acupuncture effect of "shui gou" point on amino acid neurotransmitters in extracellular fluids of striatum of acute permanent middle cerebral artery occlusion(PMCAO)model rats,it is attempted to explore time-effect rules of acupuncture effect on acute cerebral ischemia rats.Methods Microdialysis probe tube was embed on 40 healthy male SD rats.Repair 3 days,five rats were randomly allocated to blank group,the rest rats were established PMCAO models by thread embolism method that obstruct the right middle cerebral artery.Using LDF monitor PMCAO model rats’ local microcirculation blood flow changes and using Bederson criterion to NDS,Local microcirculation blood flow decreased at least 70%and NDS>2 were judged to PMCAO model were established successfully.The successfully established PMCAO rats were randomly divided into model group and acupuncture group.Acupuncture group rats will be given "shui gou" point acupuncture intervention and keeping needle 2h,consecutive acupuncture three days(the first day,the next day,the third day of PMCAO models established successfully),at the same time,using the microdialysis technique collect striatal extracellular fluids;While,the rats will not be given any intervention only microdialysis sampling under the same conditions in the seventh day.The blank and model groups’ rats will not be given any intervention only microdialysis sampling under the same conditions with the acupuncture group rats.Applying the HPLC-FLD technique to monitor the dynamic changes of amino acid neurotransmitters(Glu,GABA)from the dialysate.Results(1)Glu:Analyze the time factors:Glu in the first day of model group were increased(P<0.05),but there were no significant differences between the Glu in the next day,the third day and the seventh day of model group(P>0.05).There were no significant differences between the Glu in the acupuncture group everyday(P>0.05).There were no significant differences between the Glu in the four days of acupuncture group and model group(P>0.05).Analyze the group factors:Glu in the model and acupuncture groups were increased than the blank group in the four days and everyday(P<0.05).Glu in the acupuncture group were decreased than the model group in everyday(P<0.05).(1)GABA:Analyze the time factors:There were no significant differences between the GABA in the model and acupuncture groups everyday(P>0.05).GABA in the four days of model group were decreased(P<0.05).GABA in the four days of acupuncture group have changed(P<0.05).Analyze the group factors:GABA in the model and acupuncture groups were increased than the blank group in the four days and everyday(P<0.05).GABA in the first day of acupuncture group were decreased than the model group(P<0.05),and GABA in the next day,the third day and seventh day of acupuncture group were increased than the model group(P<0.05).Conclusion(1)"Shui gou" has a acupuncture regulating action to Glu and GABA in extracellular fluids of striatum of acute PMC AO model rats,reduce Glu excessive release caused by ischemia hypoxia,maintain endogenous GABA inhibitory effect.(2)"Shui gou" has a significant acupuncture effect to PMCAO model rats in the super early phase of acute,long time retaining acupuncture has a good stable sustained effect,and the lasting effect still exist after acupuncture in a certain stage.(3)"Shui gou" have acupuncture intervention effect,sustained effect and post effect after acupuncture in the PMCAO acute phase. |
