Objective:Observing the change of the general behavior of the rats and the area ratio of the scala media with which plus the scala vestibule,as well as the expression of AQP-2 and AQP-7 in the inner ear of endolymphatic rat by the treatment of electrical acupuncture(EA),to explore the mechanism of the electrical acupuncture for endolymphatic hydrops,providing experimental evidence for clinical treatment for Meniere’s disease.Methods:Thirty-two Wistar rats were randomly divided into a blank group,a model group,a medication group and an EA group,8 rats in each group.Model of endolymphatic hydrops were established by intraperitoneal injection of vasopressin at a dose of 50μg/kg/d for consecutive 7 days.Rats in blank group and model group were treated with identical immobilization for 20 minutes as EA group but no treatment was given;rats in medication group were treated with introgastric administration of Tolvaptan at a dose of 5μg/g/d for consecutive 10 days;rats in EA group were treated with EA at "bai hui" and left "ting gong"for 20 minutes,frequency is 2/15Hz,once a day for consecutive 10 days,then observing the indications of each group after treatment:1)behavioral changes in rats;2)Hematoxylin-Eosin staining was used to measure the severity of cochlea hydrops;3)Immunohistochemical staining was used to observe the change of AQP-2 and AQP-7 in the cochlea.Results:1)General behavioral changes:when compared with the modal group,the locomotor activity of the acupuncture group and western medicine group are increased,and the spirit is good,move smoothly,the ears reflex sensitivity,diety increases,the hair are colorful,weight increase.2)Cochlea HE staining sections showed:when compared with the blank group,model group was significantly higher in the ratio of the scala media with which plus the scala vestibule(P<0.01);when compared with the model group,western medication group and EA group were higher,but there was no significantly difference between EA group and western medicine group.3)Immunohistochemcal analysis showed:the expression of the AQP-2 and AQP-7 of the model group were upregulated than blank group(P<0.01);the expression of the AQP-2 and AQP-7 of the western medicine group and EA group were lower than modal group(P<0.01),but there was no significantly difference between EA group and western medicine group by the expression of AQP-2 and AQP-7.4)The expression of AQP-7 is higher than AQP-2 in the stria vascularis of the inner ear.Conclusions:(1)EA can relieve the endolymphatic hydrops of the Meniere’s disease rat model induced by vasopressin,the mechanism may be related to the down-regulation of theAQP-2 and AQP-7.(2)AQP-2 and AQP-7 are isotropic to regulate the formation and mitigation of the endolymphatic hydrops.
