Objective:To investigate the expression of integrin-β1 of human colon carcinoma cell line SW480 after epithelial-mesenchymal transition(EMT)and effect of invasive ability on the human colon carcinoma in vitro.Methods:TGF-β1(10ng/ml)was used to introduce EMT of SW480 cells for 72 hours.Real-time PCR and Western-blot were used to detect expression of intergin-β1 of SW480 cell.The migration of SW480 cell in vitro was assayed by transwell.Result:In contrast with control group,the expression of integrin-β1 m RNA of EMT group was increased obviously(t=3.764,P<0.05).Comparing with control group(integrin-β1/ β-actin=0.172±0.034),the expression of integrin-β1 protein of SW480 cell of EMT group(integrin-β1/β-actin=0.430±0.021)was increased obviously(P<0.05).In transwell,the number of migration of SW480 cell in EMT group(137±9)was significantly more than that in control group(36±4)(P<0.05).Conciusion:The expression of integrin-β1 and the migration of human colon carcinoma cell in vitro can be elevated after EMT.
