| Objective To establish a panel of chinese breast cancer patient-derived xenografts(PDXs)in immunodeficientmice.Methods Human breast tumor tissues were obtained from women undergoing mastectomy,lumpectomy,or core needle biopsy for a confirmed diagnosis of breast carcinoma at Ruijin Hospital.Clinical information was obtained regarding patient’s,age at diagnosis,surgical and histological pathology reports,number of positive axillary lymph nodes,and steroid receptor expression.The tissues were collected sterilely during surgery and immediately transported to our laboratory on ice in complete medium.Samples were divided into fragments(2*2mm).Fragments were implanted directly into NOD/SCID mice.Transplanted mice were monitored for tumor take and dissemination during an observation.The characteristics of the xenografted tumors and the original tumor samples were analysis.Results 239 samples of human breast cancer tumors were grafted in to NOD/SCID mice.12 tumors were taken out until now.BCKLM derived from a patient with luminal breast cancer.The latency is 14 months.BC099 derived from a patient with HER-2 positive lobular carcinoma.The latency is 3 months.Histologically,the xenografted tumor BC099 maintained the major features of the original tumor samples,but the expression of Cerb B-2 was loss.The tumor grew faster in the NOD/SCID mice than the nude mice,and the metastasis to axillary lymph node was seen in the xenograft of NOD/SCID mice.Conclusions We successfully establish a PDX.This panel represents a useful preclinical tool for testing new agents and protocols and for further exploration of the biological basis of drug responses.Our work of establishing more lines of PDXs is under way. |
