Investigation Of Mosquito And Its Infected With Viruses In Gansu Province

Background Gansu province is located in northwestern of China, with the long and narrow terrain, extending from northwest to southeast. As the development of the economy and animal husbandry in recent years, numberous mobile population is increasing, as well as the risk factors of mosquito borne diseases prevalence in Gansu province, it is necessary to investigate local mosquito and its infected virus, making out the effective diseases control measurements.7Contents On August11-26,2011and July14-19,2012, a survey for mosquito was done in Wuwei, Zhangye, Jiuquan, Lanzhou, Baiyin, Qingyangjingchang, tianshui prefectures of Gansu province, to know about the components of the mosquito. The virus was isolated by BHK-21and C6/36cell culture, and identified by the genetic test and phylogenetic trees analysis,to understand fully mosquito infected virus.Results A total of27575individual mosquitoes were collected from6species of3Genus in2subfamilies, of those mosquito species Culex pipiens pallens population quantity was the highest, its accounting for38.32%(10567/27575) of the total collected mosquitoes, as well as Aedes vexans28.70%(7914/27575), the others were Aedes flavidorsalis、Anopheles sinensis、 Culex modestus、Culex tritaeniorhynchus Respectively.Of261pools by BHK-21and C6/36cell culture isolating virus,1pool was occurred the pathological changes on both BHK-21and C6/36cells, which it was identified as Getah virus by the molecular biology tests; and35pools were only isolated from C6/36cells, of those31pools were identified as Culex Flavivirus and4Liaoning virus by molecular biology test. Getah virus came from Culex tritaeniorhynchus,31Culex Flavivirus from Culex pipiens pallens,1Liaoning virus from Aedes, and3Liaoning viruses from Culex pipiens pallens.Of261pools by RT-PCR test,67pools were the positives of viral CDNA, including the50positive products were identified as Culex Flavivirus,13as Liaoning virus and4as Getah virus.Conclusions Mosquito species were few in Gansu province, Culex pipiens pallens and Aedes vexans belonged to local predominant mosquito species, mosquito infected viruses were mainly involved in Getah virus, Culex Flavivirus and Liaoning virus, and JEV was not found in this survey. The rate of Culex pipiens pallens infected Culex Flavivirus was8.07per1000mosquitoes and0.16per1000mosquitoes for both Liaoning virus and Getah virus. Culex tritaeniorhynchus infection Getah virus rate was2.44per1000mosquitoes., and its infected Liaoning virus rate was1.22per1000mosquitoes.
